本实验用新生小牛视网膜提取物作血管内皮细胞生长促进剂,成功地使人脐静脉内皮细胞在体外连续培养23代以上。细胞倍增时间平均为16.6h,细胞传代时间平均为5 d,传代比率为1:2~1:3。用ABC法证明细胞内存在Ⅷ因子相关抗原(FⅧr-Ag),透射电镜下可见胞浆内含WP(Weibe1-Palade)小体,证实所培养的细胞为血管内皮细胞。
