人骨髓间充质干细胞向成骨细胞的诱导分化

目的诱导人骨髓间充质干细胞向成骨细胞分化。方法体外分离培养人骨髓间充质干细胞,将其分为对照组和诱导组两组。诱导组加入地塞米松、抗坏血酸和β-甘油磷酸,诱导成骨。采用免疫荧光染色的方法检测碱性磷酸酶(AKP)和骨桥蛋白(OPN)的表达;并通过RT-PCR检测骨桥蛋白mRNA的表达;利用硝酸银染色和茜素红染色两种特殊染色方法检测钙盐沉积。结果经诱导,诱导组细胞在呈典型的成骨细胞形态并有钙结节形成;免疫荧光染色结果显示诱导组细胞AKP和OPN均为强阳性表达,RT-PCR结果也显示诱导组细胞骨桥蛋白mRNA表达;
