Calf intestinal 5′-nucleotidase

Abstract The 5′-nucleotidase activity isolated from calf intestinal mucosa has been resolved into three fractions by DEAE-cellulose column chromatography. One of these fractions was obtained free of nonspecific phosphatase activity and was used for further studies.

The rate of hydrolysis of IMP (0.5 mM) by cytosol 5'-nucleotidase from Artemia embryos was increased up to 7-fold by concentrations of around 10 microM diadenosine tetraphosphate (Ap4A). Half maximal activation of the enzyme was accomplished with 5 microM Ap4A.

Nonionic, anionic and zwitterionic detergents were employed for solubilization of 5′-nucleotidase from acetone powder preparations of rat heart. Zwittergents effectively solubilized the enzyme and may allow now its further identification and purification by electrophoretic techniques.

5′-Nucleotidase in Rat Liver Lysosomes

5'-Nucleotidase was found in purified rat liver tritosomes. When tritosomes were subfractionated into the membrane and soluble contents fractions, 73% of the total 5'-nucleotidase activity was found in the membrane fraction and 24% in the soluble contents fraction.
