RPR 101821, a new potent cholesterol-lowering agent: inhibition of squalene synthase and 7-dehydrocholesterol reductase

RPR 101821 (trans-2-[4-(benzoxazol-2-yl)-phenylmethoxy] amino cyclohexane hydrochloride) is a potent cholesterol-lowering agent in rodents and marmoset. The compound inhibited rat liver microsomal squalene synthase (IC50 = 1 nM) and 7-dehydrocholesterol (7DHC) reductase (IC50 = 1 microM; Lewis et al. 1995).

Abstract 1. 1. The reduction of 7-dehydrocholesterol in cell-free rat-liver homogenates has been studied. The Michaelis constant (Km) was calculated from 11 experiments to be 1.14 · 10−3 with standard error ± 0.33 · 10−3. 2. 2. Three hypocholesterolemic agents inhibited 7-dehydrocholesterol conversion by liver homogenate.

Smith-Lemli-Opitz syndrome (SLOS) is a malformation syndrome associated with 7-dehydrocholesterol (7DHC) 7-reductase deficiency. Although SLOS can be detected in an affected fetus before midpregnancy by measurement of 7DHC levels in amniotic fluid or chorionic villus cells, a noninvasive, more routine method is needed.
