Characterizing epithelial‐mesenchymal transition‐linked heterogeneity in breast cancer circulating tumor cells at a single‐cell level

open access: yesMolecular Oncology, EarlyView.
In over 50% of non‐metastatic breast cancer patients, circulating tumor cells (CTCs) along the whole epithelial‐mesenchymal transition spectrum are detected. Total CTC number and individual phenotypes relate to aggressive disease characteristics, including lymph node involvement and higher tumor proliferation. At the single‐cell level, mesenchymal CTCs

The neural crest‐associated gene ERRFI1 is involved in melanoma progression and resistance toward targeted therapy

open access: yesMolecular Oncology, EarlyView.
ERRFI1, a neural crest (NC)‐associated gene, was upregulated in melanoma and negatively correlated with the expression of melanocytic differentiation markers and the susceptibility of melanoma cells toward BRAF inhibitors (BRAFi). Knocking down ERRFI1 significantly increased the sensitivity of melanoma cells to BRAFi.

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Non-invasive glucose prediction and classification using NIR technology with machine learning

open access: yesHeliyon
In this paper, a dual wavelength short near-infrared system is described for the detection of glucose levels. The system aims to improve the accuracy of blood glucose detection in a cost-effective and non-invasive way.

Ro 31‐8220 suppresses bladder cancer progression via enhancing autophagy in vitro and in vivo

open access: yesFEBS Open Bio, EarlyView.
The pan‐protein kinase C inhibitor Ro‐31‐8220 demonstrates potent anti‐bladder cancer effects both in vitro and in vivo by suppressing migration/invasion, inducing apoptosis and crucially activating autophagy, where blocking autophagy with chloroquine reduces its cell‐killing efficacy, suggesting its promise as a novel therapeutic candidate requiring ...

A unified model for Duchenne muscular dystrophy gene involvement in cancer: context‐dependent tumour suppression and oncogenicity

open access: yesFEBS Open Bio, EarlyView.
We propose a context‐dependent model where the Duchenne muscular dystrophy (DMD) gene acts as a tumour suppressor in aggressive tumours and as an oncogene in less aggressive ones. We propose this model as a unified framework to explain the opposing survival associations with DMD expression and to guide experimental exploration of the dual role of DMD ...

IoT Monitoring and Evaluating System for the Construction Quality of Foundation Pile

open access: yesBuildings
The quality of foundation pile is greatly influenced by human factors, and quality assessment is delayed. This paper introduces a new evaluation system based on Internet of Things (IoT) monitoring data of the foundation pile construction process.

CD4-T cell enumeration in human immunodeficiency virus (HIV)-infected patients: A laboratory performance evaluation of Muse Auto CD4/CD4% system by World Health Organization prequalification of in vitro diagnostics.

open access: yesPLoS ONE, 2019
BackgroundCD4 T-cell counts are still widely used to assess treatment eligibility and follow-up of HIV-infected patients. The World Health Organization (WHO) prequalification of in vitro diagnostics requested a manufacturer independent laboratory ...

Diffusion‐based size determination of solute particles: a method adapted for postsynaptic proteins

open access: yesFEBS Open Bio, EarlyView.
We present a diffusion‐based approach for measuring the size of macromolecules and their complexes, and demonstrate its use on postsynaptic proteins. The method requires fluorescein‐labelled protein samples, a microfluidic device that maintains laminar flow for said samples, a microscope recording the emitted fluorescent signals, and an analytic ...
