Patients in Focus: What's Relevant for Chronic Myeloid Leukaemia and Philadelphia Chromosome-Positive Acute Lymphoblastic Leukaemia?

open access: yes, 2017
This symposium was dedicated to discussing BCR-ABL-positive chronic myeloid leukaemia (CML) and Philadelphia-positive acute lymphoblastic leukaemia (Ph+ALL).

Molecular Editing Reveals a Bilobalide Chemotype that Attenuates Prolyl Endopeptidase (PREP)‐Linked Neuroinflammation

open access: yesAdvanced Science, EarlyView.
Molecular editing of bilobalide uncovers BB56, a PREP‐engaging chemotype with enhanced anti‐inflammatory activity in microglia. Integrated CETSA‐MS, SAR, and pathway analyses link BB56 to attenuation of p38 MAPK/NF‐κB and NLRP3‐associated inflammatory signaling, highlighting bilobalide editing as a strategy for developing neuroinflammation‐modulating ...

Vascularized Liver‐on‐a‐Microsphere Reveals Alanine‐Glucose Metabolism‐Driven Regulation of Liver Function and Injury

open access: yesAdvanced Science, EarlyView.
A vascularized liver‐on‐a‐microsphere system is developed to recreate lobule‐like cellular organization and endothelial barrier function within compartmentalized hydrogel particles. The platform enhances hepatic performance and reveals alanine‐glucose cycle activation linked to improved detoxification capacity, offering a high‐throughput microscale ...

Hyperpolarization of [1‐13C]Ketoisocaproate‐d2 by Reversible Exchange with Parahydrogen Enables Profiling of Branched‐Chain‐Amino‐Acid Metabolism in Cellulo and in Vivo

open access: yesAdvanced Science, EarlyView.
We report the fast generation of biocompatible solutions of highly 13C‐hyperpolarized (≈11%) partially‐deuterated ketoisocaproate within <6 min using spin‐lock‐induced crossing and reversible exchange with parahydrogen (SLIC‐SABRE). This enables rapid detection of branched‐chain‐amino‐acid transferase 1 (BCAT1) protein expression and investigation of ...

Study of the Relationship between Heme Oxygenase-1, Interleukins (6, 12) and Coenzyme Q10 in Iraqi Pediatric Patients with Chronic Myeloid Leukemia

open access: yesProceedings of the International Conference on Applied Innovations in IT
A global disorder impacted by both genetic and environmental factors, chronic myeloid leukaemia (CML) is associated with increasing mortality and morbidity rates and has social and economic consequences.

Histone H3K18 Lactylation Promotes the Malignant Progression of Wilms Tumor via a PSRC1/AKT/HIF‐1α Positive Feedback Loop

open access: yesAdvanced Science, EarlyView.
In nephroblastoma, aberrant glycolysis drives lactate accumulation, which elevates histone H3K18 lactylation via p300. Lactylation of the PSRC1 promoter activates its transcription. PSRC1 competitively binds AKT, relieving PTEN‐mediated inhibition and triggering AKT/mTOR/HIF‐1α signaling.

Exploring NK cell receptor dynamics in paediatric leukaemias: implications for immunotherapy and prognosis

open access: yesClinical & Translational Immunology
Objectives Immunotherapies targeting natural killer (NK) cell receptors have shown promise against leukaemia. Unfortunately, cancer immunosuppressive mechanisms that alter NK cell phenotype prevent such approaches from being successful.

LRG1 Drives Pathological Angiogenesis by Disrupting Neutrophil Mitochondrial Homeostasis in Bladder Cancer

open access: yesAdvanced Science, EarlyView.
In bladder cancer, LRG1 binds to ANXA2 to trigger mitochondrial ROS‐dependent NETosis. This pathogenic cascade actively uncouples endothelial‐mural cell interactions, driving profound vascular destabilization. Consequently, targeting the LRG1‐ANXA2 axis attenuates the neutrophil burden and induces structural vascular normalization, offering a powerful ...

Distinct pattern of genomic breakpoints in CML and BCR::ABL1-positive ALL: analysis of 971 patients

open access: yesMolecular Cancer
Background The BCR::ABL1 is a hallmark of chronic myeloid leukemia (CML) and is also found in acute lymphoblastic leukemia (ALL). Most genomic breaks on the BCR side occur in two regions - Major and minor - leading to p210 and p190 fusion proteins ...
