A prior status of graft‐versus‐host disease affects donor lymphocyte infusion outcomes in patients with relapsed haematological malignancies after allogeneic stem cell transplantation

open access: yesBritish Journal of Haematology, EarlyView.
Summary Optimizing the effectiveness of donor lymphocyte infusion (DLI) for relapse after allogeneic stem cell transplantation (alloHSCT) has been challenging. We investigated whether the benefits of achieving full donor chimerism (FDC) and developing graft‐versus‐host disease (GVHD) after DLI are affected by a history of GVHD before DLI.

Cancer‐Type‐Specific DNA Methylation Is a Source of Vulnerability in Liver Cancer Cells

open access: yesCancer Science, EarlyView.
We profiled genome‐wide DNA methylation in 1165 human cancer cell lines representing 25 cancer types and identified 73 genes as potential liver cancer‐specific methylation‐silenced genes. We selected ASNS, BCAT2, FXYD5, and NQO1 genes for further experimental analysis and demonstrated that aberrant methylation of BCAT2 contributes to the vulnerability ...

Ligand‐Based CAR‐T Cells Targeting EGFR Exhibit Favorable Antitumor Effects Against Gynecologic Malignancies

open access: yesCancer Science, EarlyView.
Wide variety of gynecologic malignancies express EGFR at high rates. Memory‐rich EGFR CAR‐T cells can be made by piggyBac‐mediated gene transfer with immature dendritic cells and EGFR protein. Ligand‐based EGFR CAR‐T cells have robust antitumor effects against gynecologic malignancies in vivo and in vitro.

Gastric Signet Ring Cell Carcinoma in a Helicobacter pylori‐Positive 10‐Year‐Old Boy: A Case Report and Literature Review

open access: yesCancer Science, EarlyView.
A case report and literature review of pediatric gastric signet ring cell carcinoma (GSRCC) suggest the potential role of H. pylori infection in the pathogenesis of pediatric GSRCC. ABSTRACT Gastric cancer is rare in pediatric populations, with gastric signet ring cell carcinoma (GSRCC) being an exceptionally infrequent subtype.

Targeting SHP‐1‐Mediated Inhibition of STAT3 and ERK Signalling Pathways Rescues the Hyporesponsiveness of MHC‐I‐Deficient NK‐92MI

open access: yesCell Proliferation, EarlyView.
B2M knockout (KO) represents a crucial strategy for generating universal NK‐92MI cell products. This approach results in increased phosphorylation of SHP‐1 and inhibited phosphorylation of STAT3 and ERK, which ultimately affect their cytotoxic capabilities. SHP‐1 KO facilitates the restoration of cytotoxic function in B2M KO NK‐92MI cells.

RBM39 Promotes Base Excision Repair to Facilitate the Progression of HCC by Stabilising OGG1 mRNA

open access: yesCell Proliferation, EarlyView.
Targeting RBM39 attenuates BER via destabilising OGG1 mRNA and potentiates KBrO3‐induced oxidative stress in HCC. Our study represents a valuable treatment strategy combining indisulam and an oxidative stress inducer in HCC. ABSTRACT Targeting base excision repair (BER) has been an attractive strategy in cancer therapeutics.

PI5P4K inhibitors: promising opportunities and challenges

open access: yesThe FEBS Journal, EarlyView.
Developing inhibitors is like climbing a steep, challenging mountain. PI5P4K inhibitors hold significant promise for the treatment of cancer, immune disorders, and neurological diseases. However, the path to success is fraught with hurdles, and researchers are therefore required to apply their expertise to overcome critical “checkpoints.” While some ...

Upregulated NOTCH2 Expression Is Implicated in the Clinical Aggressiveness of Atypical Fibroxanthoma and Pleomorphic Dermal Sarcoma

open access: yesInternational Journal of Dermatology, EarlyView.
ABSTRACT Background Atypical fibroxanthoma (AFX) and pleomorphic dermal sarcoma (PDS) represent clinicopathological variants of a spectrum. It is known that AFX and PDS tumors harbor frequent NOTCH1/2 mutations. However, the expression of Notch signaling pathway‐associated proteins in both tumor cells has not been studied before.
