Population Pharmacokinetics and Exposure‐Response Analyses to Support Cofetuzumab Pelidotin (ABBV‐647) Dose Optimization in Non‐Small Cell Lung Cancer Patients

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
Cofetuzumab pelidotin (Cofe‐P) is an anti‐protein tyrosine kinase 7 (PTK7) antibody‐drug conjugate (ADC) being investigated for the treatment of adults with PTK7‐expressing, recurrent non‐small cell lung cancer (NSCLC). The first‐in‐human (FIH) trial demonstrated tolerability of dosages ≤ 2.8 mg/kg once every 3 weeks (Q3W) and ≤ 3.2 mg/kg Q2W ...

Biomarkers of lung congestion and injury in acute heart failure

open access: yesESC Heart Failure, Volume 12, Issue 2, Page 781-789, April 2025.
Abstract Acute heart failure (AHF) classification and management are primarily based on lung congestion and/or hypoperfusion. The quantification of the vascular and tissue lung damage is not standard practice though biomarkers of lung injury may play a relevant role in this context.

The Serendipitous Discovery of Rollover Cyclometalation: From Unrecognized Observation to Transformative Chemistry

open access: yesEuropean Journal of Inorganic Chemistry, EarlyView.
Rollover cyclometalation is a distinctive variant of classical cyclometalation where a chelated heterocyclic ligand undergoes internal rotation, releasing one donor atom from coordination and subsequently activating a remote C─H bond with high regioselectivity.

Autonomous Nanomedicine: Closing the Biological Validation Gap

open access: yesExploration, EarlyView.
Delivery of nanoparticles to tumors has not improved in over fifteen years, and biological testing remains slow and separate from formulation design. This review proposes adding a Biology step to the design cycle, using organ chips and patient‐derived organoids, and assesses the readiness of each required technology.
