Diagnosis, Prognosis, and Drug Target Discovery for Chronic Widespread Pain: A Large Proteogenomic Study

open access: yesAdvanced Science, EarlyView.
Plasma proteomics is leveraged to decode the biological underpinnings of chronic widespread pain. A nested machine learning framework integrates proteomic signatures, prospective outcomes, and Mendelian randomization to uncover 18 causal proteins.

Tumor Intrinsic METTL5 Modulates ATF4 Translation to Prevent T Cell‐Induced Ferroptosis in Ovarian Cancer

open access: yesAdvanced Science, EarlyView.
An integrative analysis combining in vitro genome‐wide immune screens, in vivo ICB screens, and clinical data mining identifies METTL5 as a key tumor‐intrinsic immune factor. Mechanistic studies further reveal the importance of the METTL5‐ATF4‐SLC7A11/SLC3A2‐ferroptosis axis in modulating antitumor immune responses. These findings highlight METTL5 as a

Switchbody, an Antigen‐Responsive Enzyme Switch Based on Antibody and Its Working Principle

open access: yesAdvanced Science, EarlyView.
This paper presents an antibody‐based enzyme switch, termed a “Switchbody”, which is fused with an enzyme‐derived fragment to the N‐terminus of an antibody. As its working principle, the fused fragment is trapped within the antibody and released upon antigen binding, leading to enzyme reconstitution and a subsequent increase in enzyme activity.

Acetylation of the Mitochondrial Chaperone GRP75 Governs ER‐Mitochondrial Calcium Homeostasis and Hepatocyte Insulin Resistance

open access: yesAdvanced Science, EarlyView.
Overnutrition exacerbates insulin resistance (IR) and is linked to excessive mitochondrial protein acetylation. However, it remains unclear whether and how mitochondrial protein acetylation influences IR. GCN5L1‐dependent downregulation of GRP75 acetylation at K567/612 consequently enhances ER‐mitochondrial calcium flux and induces ER stress.
