Inhibition of CDK9 enhances AML cell death induced by combined venetoclax and azacitidine

The CDK9 inhibitor AZD4573 downregulates c‐MYC and MCL‐1 to induce death of cytarabine (AraC)‐resistant AML cells. This enhances VEN + AZA‐induced cell death significantly more than any combination of two of the three drugs in AraC‐resistant AML cells.

Purification and Characteristics of .BETA.-Galactosidase from Alternaria alternate.

Alt. alternateを小麦ふすま培地に培養し, β-ガラクトシダーゼ粗酵素液を抽出した.硫酸アンモニウム塩析の後, Sephacryl S-200, DEAE-TOYOPEARL 650MおよびHydroxyapatiteカラムクロマトグラフィーを順次用いて精製を行った.その結果,比活性で174倍に精製され,回収率は23.6%であった.この段階で酵素は活性染色では単一のバンドを示したが,単一タンパク質を得るまでには至らなかった.この部分精製酵素の分子量は125000,最適反応pHはONPGを基質とした場合4.0~4.5,ラクトースを基質とした場合5.0~5.5で, pH 4.0~8.5の範囲内で安定であった.最適反応温度は両基質に対しても60℃であり, pH 4.5・1時間保持においては ...

Adaptaquin is selectively toxic to glioma stem cells through disruption of iron and cholesterol metabolism

Adaptaquin selectively kills glioma stem cells while sparing differentiated brain cells. Transcriptomic and proteomic analyses show Adaptaquin disrupts iron and cholesterol homeostasis, with iron chelation amplifying cytotoxicity via cholesterol depletion, mitochondrial dysfunction, and elevated reactive oxygen species.

Exploring $0\nu\beta\beta$ and Leptogenesis in the Alternative Left-Right Model

We investigate the possibility of neutrinoless double beta decay ($0\nu\beta\beta$) and leptogenesis within the Alternative Left-Right Model (ALRM). Unlike the usual left-right symmetric model, ALRM features a Majorana right-handed neutrino which does not carry any charge.

The neural crest‐associated gene ERRFI1 is involved in melanoma progression and resistance toward targeted therapy

ERRFI1, a neural crest (NC)‐associated gene, was upregulated in melanoma and negatively correlated with the expression of melanocytic differentiation markers and the susceptibility of melanoma cells toward BRAF inhibitors (BRAFi). Knocking down ERRFI1 significantly increased the sensitivity of melanoma cells to BRAFi.

Survivin and Aurora Kinase A control cell fate decisions during mitosis

Aurora A interacts with survivin during mitosis and regulates its centromeric role. Loss of Aurora A activity mislocalises survivin, the CPC and BubR1, leading to disruption of the spindle checkpoint and triggering premature mitotic exit, which we refer to as ‘mitotic slippage’.

CDK11 inhibition induces cytoplasmic p21WAF1 splice variant by p53 stabilisation and SF3B1 inactivation

CDK11 inhibition stabilises the tumour suppressor p53 and triggers the production of an alternative p21WAF1 splice variant p21L, through the inactivation of the spliceosomal protein SF3B1. Unlike the canonical p21WAF1 protein, p21L is localised in the cytoplasm and has reduced cell cycle‐blocking activity.
