Evolutionarily divergent DUF4465 domains have a common vitamin B12‐binding function

open access: yesFEBS Open Bio, EarlyView.
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Mass spectrometry based identification of AMP‐O‐Tris generated by Thermococcus onnurineus Cas10

open access: yesFEBS Open Bio, EarlyView.
Isolated Thermococcus onnurineus Cas10 generates the noncanonical ATP‐derived product AMP‐O‐Tris while in Tris‐containing buffer as identified via mass spectrometry, revealing relaxed nucleophile selectivity under isolated conditions. These findings suggest that multiprotein Csm complex assembly restricts Cas10 reactivity toward canonical cyclic ...

Pitaya as a New Alternative Crop for Iberian Peninsula: Cultural Practices

open access: yesPlants
Pitaya (Selenicereus spp.) cultivation has expanded in the Iberian Peninsula in recent years, driven mainly by increasing demand from the European market and by the crop’s good adaptability to Mediterranean conditions.

Long‐term hippocampal alterations and cognitive impairment in a murine model of surgical sepsis

open access: yesFEBS Open Bio, EarlyView.
Using a mouse model of surgical sepsis, we tested long‐term memory and analyzed the transcriptome of single cells isolated from the hippocampus. Survivor mice showed worse memory, loss of certain brain cell subpopulations, and abnormal immune cell activity—suggesting that post‐sepsis brain alterations may be linked to cognitive deficits.

Identifying gene expression signatures for risk stratification of postoperative adjuvant chemotherapy in colorectal cancer

open access: yesFEBS Open Bio, EarlyView.
A novel signature integrating genome‐wide analysis with clinical factors predicts recurrence in stage II colorectal cancer and enables a new risk stratification to guide postoperative adjuvant chemotherapy. Clinical risk stratification for postoperative recurrence in patients with pathological stage II (pStage II) colorectal cancer (CRC) is essential ...

Effects of replacing rye silage with mixed rye-vetch-straw silage on feed intake, milk production, digestion processes, and blood metabolites in dairy cows

open access: yesJournal of Dairy Science
: Mixed silage of whole catch crops—crops grown between 2 main crops—and straw might be an alternative forage source for dairy cattle in times of forage shortage but is still understudied. This research sought to evaluate the effects of feeding mixed rye-

YIPFα1A expression is regulated by multilayered molecular mechanisms

open access: yesFEBS Open Bio, EarlyView.
YIPFα1A, a five‐pass Golgi protein, is regulated at multiple layers. (1) Rare‐codon enrichment drives translation‐coupled mRNA decay. (2) A proximal 3′‐UTR element stabilizes mRNA. (3) A distal 3′‐UTR element included by alternate poly(A) site usage represses translation, which can be overridden by the proximal 3′‐UTR element.

Performance, feed-sorting behavior, methane and carbon dioxide emissions, nitrogen balance, and nutrient digestibility of dairy heifers fed Kernza intermediate wheatgrass straw

open access: yesJournal of Dairy Science
: Kernza straw (KS) is a high-fiber, low-energy byproduct of intermediate wheatgrass (Thinopyrum intermedium [Host] Barkworth & D.R. Dewey) that may serve as an alternative forage to increase dietary fill and reduce the risk of over-conditioning in ...
