The single-dose pharmacokinetics of amiodarone have been studied in volunteer subjects given 400 mg doses by the intravenous and oral routes. The data show the compound to have a very large volume of distribution, a low total clearance, and a long and variable terminal elimination half-life.

Amiodarone, a drug used to treat refractory cardiac arrhythmias, produced a peripheral neuropathy in 5 of 50 cases (10%). Although the neuropathy may be severe, it tends to improve with lowering of the dosage or discontinuation of the medication.

6 cardiac patients were found to present various degrees of corneal involvement following systemic treatment with amiodarone. The clinical pattern of the keratopathy, its benign course and the pathophysiology are discussed. Lacrimal insufficiency or incomplete blinking seem to contribute to the severity of the keratopathy.

Assessment of TSH and TPO-Ab before starting amiodarone (AM) treatment is recommended. The usefulness of periodic TSH measurement every 6 months during AM treatment is limited by the often sudden explosive onset of AIT, and the spontaneous return of a suppressed TSH to normal values in half of the cases.

Abstract Amiodarone is a potent new antiarrhythmic agent that is structurally similar to thyroxine. It has a long half-life (about 1 month), which makes management of side effects difficult. It crosses the placenta, so its use in women of child-bearing age is a problem in terms of possible fetal damage, even if it is stopped before conception ...

ABSTRACT.Amiodarone has proved very effective in the treatment of otherwise resistant cardiac tachyarrhythmias. The use of amiodarone has, however, been limited due to its serious side‐effects. A patient with cholestatic hepatitis due to amiodarone treatment is presented below and a review of the hepatotoxicity of amiodarone is given.
