Employing directed evolution, we modified AlkS to detect isobutanol, significantly improving biosensor specificity. Subsequent optimisations through site‐directed mutagenesis and promoter engineering further enhanced the dynamic range to 5.56‐fold change, equivalent to a 114% increase.
