Single‐Cell Dissection of Therapy‐Induced Remodeling Uncovers a Fibroblast‐Driven Immunosuppressive Niche and Targetable Vulnerabilities in Lethal Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Single‐cell longitudinal profiling reveals that androgen‐deprivation therapy induces a DPT+ fibroblast‐complement axis that suppresses macrophage inflammation and drives CD8+ T cell exhaustion in prostate cancer. Concurrently, resistant epithelial subpopulations persist and engage TSPAN1‐ and NRXN1‐mediated programs promoting CRPC and neuroendocrine ...

A Recurrent Germline Mutation in the 5'UTR of the Androgen Receptor Causes Complete Androgen Insensitivity by Activating Aberrant uORF Translation. [PDF]

open access: yesPLoS ONE, 2016
A subset of patients with monogenic disorders lacks disease causing mutations in the protein coding region of the corresponding gene. Here we describe a recurrent germline mutation found in two unrelated patients with complete androgen insensitivity ...

EP300‐Mediated MTF1‐K218 Lactylation Buffers AR‐Driven Copper Overload to Suppress Cuproptosis in Castration‐Resistant Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Androgen receptor (AR) drives copper accumulation in prostate cancer while inducing MTF1 to buffer copper toxicity. EP300‐mediated lactylation of MTF1 at K218 promotes its nuclear translocation and metallothionein expression, sequestering cytosolic copper and preventing mitochondrial cuproptosis.

SAV1 in Cooperation With FKBP52 Participates in Implantation

open access: yesAdvanced Science, EarlyView.
The MST‐SAV1 complex modulates FKBP52 phosphorylation to fine‐tune PR activity, ensuring timely uterine transition to receptivity under estrogen signaling for implantation. Stromal SAV1 scaffolds MST1/2‐mediated FKBP52 phosphorylation via noncanonical Hippo to promote PR activity.

Human biomarker navigator

open access: yesiMeta, EarlyView.
The Human Biomarker Navigator integrates the disease continuum, biomarker dynamics, cross‐organ biomarker networks, biomarker classification, and technology‐driven paradigms. It maps how biomarkers link multi‐system physiology and pathology across the nervous, respiratory, endocrine, circulatory, immune, digestive, urinary, reproductive, and ...

Androgen insensitivity syndrome: discussion based on three cases [PDF]

open access: yes, 2017
Androgen insensitivity syndrome is an X-linked recessive condition due to a complete or partial insensitivity to androgen, resulting in a failure of normal masculinisation of the external genitalia in chromosomally male individuals.

Human Prostate Cell Lines: Derivation, Molecular Characteristics and Appropriate Use

open access: yesThe Prostate, EarlyView.
ABSTRACT Background Research on prostate cancer and benign prostatic hyperplasia depends on a small number of immortalized human cell lines established between 1977 and 2001, most of them from advanced metastatic disease. The cell lines differ from one another in androgen receptor status, prostate‐specific antigen secretion, p53 status, karyotype and ...

The androgen receptor and signal-transduction pathways in hormone-refractory prostate cancer. Part 1: modifications to the androgen receptor [PDF]

open access: yes, 2005
Prostate cancer is the second most common male malignancy in the western world an increasing incidence in an ageing population. Treatment of advanced prostate cancer relies on androgen deprivation.

Evaluating the Term ‘Disorders of Sex Development’: A Multidisciplinary Debate

open access: yesSocial Medicine, 2018
In 2014, almost 10 years after the 2005 International Consensus Conference on Intersex in Chicago, one of the conference co-organisers, under the auspices of a number of international paediatric endocrinology societies, launched the Global DSD Update to ...

Challenges in clinical and laboratory diagnosis of androgen insensitivity syndrome: a case report

open access: yesJournal of Medical Case Reports, 2011
Introduction Androgen is a generic term usually applied to describe a group of sex steroid hormones. Androgens are responsible for male sex differentiation during embryogenesis at the sixth or seventh week of gestation, triggering the development of the ...
