Key Connectomes and Synaptic‐Compartment‐Specific Risk Genes Drive Pathological α‐Synuclein Spreading

Utilizing a stereotaxic injection mouse model and a novel mathematical approach, this study uncovers key subnetworks that drive pathological α‐synuclein (α‐Syn) progression in Parkinson's disease (PD). Remarkably, just 2% of the strongest connections in the connectome are sufficient to predict its spread.

Gallic acid (GA) and copper ions self‐assemble to form nanoparticles, which are then modified with mitochondrial targeting peptides and gap junction modulator. These nanoparticles scavenge mitochondrial reactive oxygen species to induce M2 polarization and enhance intercellular mitochondrial transfer.

The P6CIT lipopolymer demonstrates superior transfection efficacy in the spinal cord and DRG, as well as enhanced penetration of the pia mater compared to the MC3 ionizable lipid. Intrathecal delivery of P6CIT LPNPs enables effective transfection in the gliocytes in the spinal cord and DRG.
