Polycarbonate nanogels enable lipid‐free delivery of anti‐MFAP‐5 siRNA into cancer‐associated fibroblasts (CAF) in hepatocellular carcinoma. In a cirrhotic murine model, this approach silences MFAP‐5, reduces fibroblast activation, and suppresses tumor growth by inhibiting NOTCH/Hes1‐driven angiogenesis. A Systemic Selective Modified mRNA Delivery Platform for Preventing Chemotherapy‐Induced Cardiotoxicity

In Yoo and Mainkar et al., we present a minimally invasive, CM‐selective modRNA delivery system encapsulated in lipid nanoparticles for intravenous (IV) administration. This platform enables selective cardiac translation of therapeutic modRNA but suppresses expression in off‐target tissues, including tumors.

Biohybrid Tendons Enhance the Power‐to‐Weight Ratio and Modularity of Muscle‐Powered Robots

Biohybrid robots powered by skeletal muscle actuators are capable of dynamically adapting to environmental cues. This study takes inspiration from native muscle–tendon architecture by leveraging tough hydrogels as synthetic tendons for muscle actuators to enhance the power‐to‐weight ratio and modularity of biohybrid machines.

Quantitative Proteomics and CRISPR/Cas9 Editing Reveal UPR‐Mediated Control of Immunoglobulin Homeostasis in Hybridomas

BCR sequencing and subclone analysis correlated immunoglobulin (Ig) chain loss in dysfunctional hybridomas with disrupted monoclonal antibody homeostasis. Proteomics‐guided CRISPR/Cas9 editing revealed that the unfolded protein response (UPR) regulates aberrant Ig synthesis.

Advanced Human Immune Cell‐Organoid Co‐Cultures for Functional Testing of Cancer Nanovaccines

Pancreatic ductal adenocarcinoma remains difficult to treat. We establish an organoid–immune co‐culture using patient‐derived organoids and matched T‐cells to assess cancer vaccines. A mesothelin‐targeted nanovaccine activates antigen‐specific T‐cells, increases IFN‐γ, and targets MSLN+ organoids.
