Organ‐specific redox imbalances in spinal muscular atrophy mice are partially rescued by SMN antisense oligonucleotides

open access: yesFEBS Letters, EarlyView.
We identified a systemic, progressive loss of protein S‐glutathionylation—detected by nonreducing western blotting—alongside dysregulation of glutathione‐cycle enzymes in both neuronal and peripheral tissues of Taiwanese SMA mice. These alterations were partially rescued by SMN antisense oligonucleotide therapy, revealing persistent redox imbalance as ...

Transferrin receptor 1‐mediated iron uptake supports thermogenic activation in human cervical‐derived adipocytes

open access: yesFEBS Letters, EarlyView.
In this study, we found that human cervical‐derived adipocytes maintain intracellular iron level by regulating the expression of iron transport‐related proteins during adrenergic stimulation. Melanotransferrin is predicted to interact with transferrin receptor 1 based on in silico analysis.

Automata and Answer Set Programming [PDF]

open access: yes, 2008
In answer set programming (ASP), one does not allow the use of function symbols. Disallowing function symbols avoids the problem of having logic programs which have stable models of excessively high complexity. For example, Marek, Nerode, and Remmel showed that there exist finite predicate logic programs which have stable models but which have no ...

Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Answer Set Programming in a Nutshell

open access: yesEPiC Series in Computing, 2018
Answer Set Programming (ASP) has emerged in the recent years as a powerful paradigm for declarative problem solving, which has its roots in knowledge representation and non-monotonic logic programming. Similar to SAT solving, the basic idea is to encode solutions to a problem in the models of a non-monotonic logic program, which can be computed by ...

Modelling Grammar Constraints with Answer Set Programming [PDF]

open access: yes, 2011
Representing and solving constraint satisfaction problems is one of the challenges of artificial intelligence. In this paper, we present answer set programming (ASP) models for an important and very general class of constraints, including all constraints

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

Valosin‐containing protein counteracts ATP‐driven dissolution of FUS condensates through its ATPase activity in vitro

open access: yesFEBS Letters, EarlyView.
Biomolecular condensates formed by fused in sarcoma (FUS) are dissolved by high ATP concentrations yet persist in cells. Using a reconstituted system, we demonstrate that valosin‐containing protein (VCP), an AAA+ ATPase, counteracts ATP‐driven dissolution of FUS condensates through its D2 ATPase activity.

A Theoretical Bound Which Improves the Performance of Compilation-Based Multi-Agent Path Finding

open access: yesIEEE Access
A well-known approach to optimally solving Multi-Agent Path Finding (MAPF) is by compilation to Boolean Satisfiability or Answer Set Programming. Such compilation-based approaches to MAPF are superior to others on dense, relatively small instances ...

ASP-based method for the enumeration of attractors in non-deterministic synchronous and asynchronous multi-valued networks

open access: yesAlgorithms for Molecular Biology, 2017
Background This paper addresses the problem of finding attractors in biological regulatory networks. We focus here on non-deterministic synchronous and asynchronous multi-valued networks, modeled using automata networks (AN).
