Expression of ARF6 mutants in neuroendocrine cells suggests a role for ARF6 in synaptic vesicle biogenesis [PDF]

open access: yesFEBS Letters, 2001
ARF6 regulates membrane trafficking between the plasma membrane and endosomes. We investigated the role of ARF6 in synaptic vesicle biogenesis as this process occurs both at the plasma membrane and at endosomes. We used a synaptic vesicle marker protein,

Arf6 regulates energy metabolism in neutrophils

open access: yesFree Radical Biology and Medicine, 2021
The small GTPase Arf6 regulates many cellular processes, including cytoskeletal remodeling, receptor endocytosis, and pathogen phagocytosis. Arf6 silencing in neutrophil (PMN)-like cells is well-known to inhibit chemotactic peptide-mediated activation of phospholipase D, the oxidative burst, and β2 integrin-dependent adhesion.

ARF6 activation by Gαq signaling: Gαq forms molecular complexes with ARNO and ARF6

Cellular Signalling, 2006
G protein-coupled receptors (GPCRs) are widely expressed hepta-helical receptors with tightly regulated pleiotropic effects. ADP-Ribosylation Factor 6 (ARF6) plays an important role in GPCR trafficking and is the subject of intense research. However, the mechanisms underlying activation and regulation of ARF6 by GPCRs are poorly characterized.

Actin Assembly at Membranes Controlled by ARF6 [PDF]

open access: yesTraffic, 2000
The small GTPase, ADP‐ribosylation factor‐6 (ARF6), has been implicated in regulating membrane traffic and remodeling cortical F‐actin. Using real‐time video analysis of actin assembly in living cells, we investigated the function and mechanism of ARF6 in control of actin assembly. Expression of an activated form of ARF6 that mimicks the GTP‐bound form

Localization of endogenous ARF6 to sites of cortical actin rearrangement and involvement of ARF6 in cell spreading

Journal of Cell Science, 1998
ABSTRACT To study the function of the endogenous ARF6 GTP binding protein in cells, we generated an antibody which specifically recognizes ARF6, and not the other ARF proteins. Using this antibody, ARF6 was detected in all mouse organs tested and in a variety of cultured cell lines including RBL, MDCK, NRK, BHK, COS, and HeLa cells.

Activation machinery of the small GTPase Arf6

Advances in Biological Regulation, 2014
The small GTPase ADP-ribosylation factor 6 (Arf6) plays pivotal roles in a wide variety of cellular events, including exocytosis, endocytosis, actin cytoskeleton reorganization and phosphoinositide metabolism, in various types of cells. To control such a wide variety of actions of Arf6, activation of Arf6 could be precisely controlled by its activators,

Preparation of Myristoylated Arf1 and Arf6

2005
Arf proteins are members of the Arf family of small Ras-like GTP binding proteins. Six Arfs, grouped into three classes, have been identified in mammalian cells and three members have been identified in yeasts. Arf1 and Arf6, more extensively studied than other Arfs, have been found to affect membrane traffic and actin remodeling.

Localization and regulation of phospholipase D2 by ARF6

Journal of Cellular Biochemistry, 2005
AbstractPhospholipase D (PLD) and ADP‐ribosylation factor 6 (ARF6) have been implicated in vesicular trafficking and rearrangement of the actin cytoskeleton. We have explored the co‐localization of rat PLD1b and rat PLD2 with wild type and mutant forms of ARF6 in HeLa cells and studied their activation by ARF6 and the role of the actin cytoskeleton ...

A requirement for ARF6 during the completion of cytokinesis

Experimental Cell Research, 2005
During cancer development, coordinated changes in cell motility and cell cycle progression are required for the gradual transformation of normal cells into cancer cells. Previous studies have shown that ARF6 is a critical regulator of epithelial cell integrity and motility via its role in membrane movement and actin-based cytoskeletal remodeling ...
