ATP-binding cassette transporter A1 and cholesterol trafficking
Current Opinion in Lipidology, 2002Two hallmarks of cardiovascular disease are the presence of sterol-laden macrophages in the artery wall and reduced plasma HDL levels. A cell membrane protein named ATP-binding cassette transporter A1 (ABCA1) mediates secretion of excess cholesterol from cells into the HDL metabolic pathway.

The appearance of scavenger receptor class B type I (SR-BI) and ATP-binding cassette transporter A1 (ABCA1) in macrophages and liver implicates these transporters in different stages of reverse cholesterol transport. This review focuses on the role of SR-BI and ABCA1 in reverse cholesterol transport in the context of atherosclerotic lesion development ...

Dynamic regulation of alternative ATP-binding cassette transporter A1 transcripts
Biochemical and Biophysical Research Communications, 2003The ATP-binding cassette transporter A1 (ABCA1) gene, a major regulator of high-density lipoprotein (HDL) metabolism, utilizes several transcriptional start sites. The pattern of transcripts among tissues and cells differed significantly. The longest (class 1) transcripts were abundant in adult brain and fetal tissues.

Statins downregulate ATP-binding-cassette transporter A1 gene expression in macrophages
Biochemical and Biophysical Research Communications, 2004The ATP-binding-cassette transporter A1 (ABCA1) plays an essential role in cellular cholesterol efflux and helps prevent macrophages from becoming foam cells. The statins are widely used as cholesterol-lowering agents and have other anti-atherogenic actions.

ATP-binding cassette transporter A1: regulation of cholesterol efflux
Biochemical Society Transactions, 2004The ATP-binding cassette transporter A1 (ABCA1) is involved in the regulation of cholesterol efflux from cells. Mutations in ABCA1 give rise to familial high-density lipoprotein (HDL) deficiency and Tangier disease, which is characterized by very low levels of HDL in plasma and cholesteryl ester accumulation in tonsils and other reticuloendothelial ...

Regulation of ATP-Binding Cassette Transporter A1 Transcription by Thyroid Hormone Receptor
Biochemistry, 2004Transcriptional regulation of the ATP-binding cassette transporter (ABCA1) gene is complex. It involves multiple transcription start sites and the binding of several different transcription factors to the ABCA1 promoter region. Cholesterol- and oxysterol-mediated up-regulation of ABCA1 transcription includes the binding of the liver X receptor and ...

ATP-binding cassette transporter A1, fatty acids, and cholesterol absorption
Current Opinion in Lipidology, 2003A significant advance in our understanding of the reverse cholesterol transport pathway occurred in 1999 with the identification of defects in the ATP-binding cassette transporter A1 gene as the cause of Tangier disease. Since this discovery, an overwhelming number of experiments have been conducted to further define the function of this gene.

ATP-Binding cassette transporter A1 is involved in hepatic α-tocopherol secretion
The Journal of Nutritional Biochemistry, 2010Vitamin E (alpha-tocopherol) is an essential fat-soluble nutrient with antioxidant properties. alpha-Tocopherol transfer protein (alpha-TTP), the product of the gene responsible for familial isolated vitamin E deficiency, plays an important role in maintaining the plasma alpha-tocopherol level by mediating the secretion of alpha-tocopherol by the liver.

Effects of Pravastatin on the Expression of ATP-Binding Cassette Transporter A1
The Journal of Pharmacology and Experimental Therapeutics, 2004In vitro inhibition of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase causes the suppression of liver X receptor (LXR) activity. Because LXR regulates the expression of ATP-binding cassette transporter (ABC) A1, which is involved in the high-density lipoprotein-related reverse cholesterol transport pathway, we examined the effects of an HMG ...
