PFKFB4 Deubiquitination by USP10 Enhances Fumarate Metabolism to Orchestrate the KDM1A/Rad51 Axis and Confer Radioresistance in Lung Cancer

open access: yesAdvanced Science, EarlyView.
USP10 binds to and stabilizes PFKFB4, enhancing glycolytic ATP production, which activates the urea cycle and elevates fumarate. This inhibits histone demethylase KDM1A, leading to increased H3K4me1 enrichment at the Rad51 promoter and direct activation of Rad51 transcription, which confers lung cancer radioresistance. The PFKFB4 inhibitor 5MPN targets

EphB1‐Mediated Transient Blood‐Brain Barrier Opening Facilitates a Ferritin‐Based Nanotherapeutic for Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
We constructed a brain‐targeted AFn‐DPZ nanodrug delivery system and identified EphB1 as its receptor on the blood‐brain barrier (BBB). This system facilitates drug transport across the BBB and thereby ameliorates cognitive impairment in Alzheimer's disease.

Somatic Evolution of a Germline Antibody Expands its Breadth to Neutralize Early SARS‐CoV‐2 Omicron Variants

open access: yesAdvanced Science, EarlyView.
Engineering of the 148‐germline (148‐GL) antibody reveals a roadmap for overcoming SARS‐CoV‐2 viral escape. By introducing four specific somatic hypermutations, the resulting 148‐M4 variant restores potent neutralization against Omicron BA.1 and BA.4/5.

Wearable Electro‐Thermal Haptic Stimulator Driven by a Self‐Powered Tactile Sensor for Realistic Stimulus Replication

open access: yesAdvanced Science, EarlyView.
We present a stimulus‐replicating system combining a tactile sensor with a wearable Peltier thermotactile module and concentric electrotactile electrode. Signals encoding pressure and temperature drive the stimulators to reproduce mechano‐thermal cues on the skin. AlN/PDMS encapsulation and SA/LA PSA ensure mechanical compliance and thermal efficiency.
