Reconstitution of autophagosome nucleation defines Atg9 vesicles as seeds for membrane formation

open access: yesScience, 2020
Reconstituting autophagosome nucleation To stay healthy, our cells must constantly dispose of harmful material. Autophagy, or self-eating, is an important mechanism to ensure the clearance of bulky material.

ESCRT‐Mimetic Nanodegrader Targets STING for Anti‐Inflammatory Therapy

open access: yesAdvanced Science, EarlyView.
A nanoplatform‐enabled targeted protein degradation strategy is presented to regulate aberrant STING signaling. STING‐ATTEC induces selective autophagic degradation of STING via formation of a STING–ATTEC–LC3 ternary complex, while the cationic FA‐LNP+ system enhances LC3 generation and targeted delivery. Together, this synergistic approach efficiently

TP53INP2 contributes to autophagosome formation by promoting LC3-ATG7 interaction

open access: yesAutophagy, 2019
TP53INP2/DOR (tumor protein p53-inducible nuclear protein 2) contributes to mammalian macroautophagy/autophagy by carrying nuclear deacetylated MAP1LC3/LC3 to the cytoplasm.

Endogenous Engineering Reprograms Extracellular Vesicles for Enhanced Therapeutic Function

open access: yesAdvanced Science, EarlyView.
This review explains how Extracellular vesicles‐producing cells can be endogenously engineered to load therapeutic proteins and nucleic acids. We summarize physiological and genetic strategies that harness native sorting pathways for selective cargo loading.

Decoding the Mechanism of Action of a Parasite TGFβ Antagonist Inspires the Creation of Cell‐Type‐Specific TGFβ Modulators

open access: yesAdvanced Science, EarlyView.
The mammalian TGFβ interacts with ubiquitously expressed TGFBR1 and TGFBR2, and current TGFβ‐targeting agents are non‐cell‐selective. The cooperative interaction of the modular parasite TGFβ antagonist with multiple host (co‐)receptors empowers the design of TGM chimeras and bispecific antibodies that activate or inhibit TGFβ signaling in a cell ...

A Biomimetic Nanoparticle System Intercepts and Degrades Thrombospondin‐1 to Restore Vascular Homeostasis After Ischemic Injury

open access: yesAdvanced Science, EarlyView.
TAX2‐NPs capture extracellular TSP‐1 in the injured liver and promote its macrophage‐mediated autophagic degradation. This process blocks TSP‐1/CD47 signaling, restores VEGFR2‐AKT activity, preserves endothelial function, and mitigates hepatic ischemia‐reperfusion injury.

Vac8 spatially confines autophagosome formation at the vacuole in S. cerevisiae

open access: yesJournal of Cell Science, 2019
Autophagy is initiated by the formation of a phagophore assembly site (PAS), the precursor of autophagosomes. In mammals, autophagosome formation sites form throughout the cytosol in specialized subdomains of the endoplasmic reticulum (ER). In yeast, the

Targeting Endogenous Lipophagy: A Novel Strategy to Enhance MSC Osteogenesis and Mineralization for Senile Osteoporosis Therapy

open access: yesAdvanced Science, EarlyView.
Schematic representation of the role of lipophagy in bone mesenchymal stem cells(MSCs). In healthy MSCs, functional lipophagy efficiently degrades lipid droplets to support oxidative phosphorylation and cellular energy production, thereby facilitating osteogenic differentiation and matrix mineralization.

BCAS-3 is required for the progression of autophagosome formation to degrade paternal mitochondria in Caenorhabditis elegans

open access: yesiScience
Summary: In the nematode Caenorhabditis elegans, autophagy degrades paternal mitochondria after fertilization to ensure the maternal inheritance of mitochondrial DNA.

Atg14: A Key Player in Orchestrating Autophagy

open access: yesInternational Journal of Cell Biology, 2011
Phosphorylation of phosphatidylinositol (PtdIns) by a PtdIns 3-kinase is an essential process in autophagy. Atg14, a specific subunit of one of the PtdIns 3-kinase complexes, targets the complex to the probable site of autophagosome formation, thereby ...
