Covalent drug discovery: Progress against key targets, emerging strategies and lessons learnt
Abstract Covalent drug discovery is currently experiencing a boom in industrial and academic interest. To date, at least 75 covalent drugs have received regulatory approval, targeting both traditional target classes and more challenging proteins for which other approaches failed. In many cases, unique aspects of covalent targeting are essential for the

Mice expressing only mutant PTEN Y138L, a protein which shows normal suppression of cellular AKT yet lacks protein phosphatase activity, die in utero, and heterozygous mice display a range of tumors. This indicates both the lipid and protein phosphatase activities of PTEN work together for normal tumor suppression and embryonic development.

Blocking protein quality control degradation by deleting the E3 ligases Ubr1 or San1 stabilizes many DHFR insertion and deletion variants that are otherwise partially unfolded. These hypomorphic variants regain folding at lower temperatures or upon methotrexate binding, revealing that PQC degradation can remove proteins that are still functionally ...

TOP1MT rs2293925 is an enhancer‐active regulatory SNP that shapes mitochondrial R‐loop dynamics
This study shows how a common genetic variant of mitochondrial topoisomerase 1 (TOP1MT rs2293925) can influence mitochondrial gene regulation, DNA topology, and formation of noncanonical nucleic acid structures such as R‐loops. By linking this enhancer‐active variant to mitochondrial nucleic acid stress in cellular contexts relevant to amyotrophic ...

ABSTRACT As a periodization that is generally understood to refer to an era of Terrestrial relationships that developed with modernization and nuclearization, the Anthropocene cannot be applied simply to premodern histories and texts. However, it has introduced historians to geological periodization as a mode of describing periods of changed ...

A review of skin microbiome and new challenges to cosmetic microbiome‐friendly formulations
Microbiome‐friendly concepts offer new perspectives for a healthy relationship between cosmetic use and the skin microbiome, especially the interaction of non‐biotic cosmetic ingredients with the skin, requiring further studies to clarify the dynamics between the skin microbiome and different classes of cosmetic ingredients.

Expert Asymmetry: Evidence From Securities Litigation
ABSTRACT Modern litigation often involves two separate, extra‐legal features: (1) contingency fee arrangements with the plaintiff‐side attorney, and (2) a “battle of the experts” where the outcome of the case rests on conflicting expert witness testimony.

Abstract Giant kelp (Macrocystis pyrifera) forests have declined by 95% in Australia since the 1970s due to climate change. Despite expanding restoration efforts, long‐term recovery is still lacking. Factors influencing recruitment by the microscopic gametophyte phase may represent a key mechanism underpinning the successful recovery and restoration of
