The selective activation of a relatively non-toxic prodrug by an enzyme present only in the tumour should enhance the drug concentration at the tumour site and result in a better anti-tumour effect and a reduction in systemic toxicity as compared to conventional chemotherapy. beta-Glucuronidase is such an enzyme.

Beta-glucuronidase in human cutaneous tumours

Abstract β-glucuronidase activity in human malignant melanomas ( 34 cases) is compared with other benign and malignant pigmentary or non-pigmentary cutaneous tumours. The results show good agreement between histochemical and biochemical study and indicate a statistically significant rise of β-glucuronidase activity in human malignant melanomas.

Beta-glucuronidase activity has been investigated in 1,221 Enterobacteriaceae strains. The test, using p-nitrophenyl-beta-D-glucuronide (pNPGlcU) agar culture, can be included easily in the routine Enterobacteriaceae biochemical assays.
