Biginelli Reaction: A Green Perspective
Current Organic Chemistry, 2012The Biginelli Reaction is a one-pot acid catalysed cyclocondensation of -keto ester, urea and aromatic aldehyde which leads to the synthesis of functionalised 3,4-dihydro-2(H)-pyrimidinones (DHPMs). This three-component reaction for the synthesis of dihydro- pyrimidinone and corresponding dihydropyrimidinethiones has now been known for more than a ...

2-Acylthioacetamides in the Biginelli Reaction
Chemistry of Heterocyclic Compounds, 2014We have demonstrated for the first time a Biginelli reaction of 2-acylthioacetamides with aromatic aldehydes and ureas or thioureas, leading to N-Ar1-4-Ar2-6-R1-1-R2-2-oxo(thioxo)-1,2,3,4-tetrahydro-pyrimidine-5-carbothioamides. The regioselectivity of this process matched the concept of hard/soft Lewis acids and bases.

Multicomponent Combinatorial Polymerization via the Biginelli Reaction
Journal of the American Chemical Society, 2016A multicomponent combinatorial polymerization method has been exploited as a new intersection between combinatorial chemistry, polymer chemistry, and organic chemistry. The tricomponent Biginelli reaction has been employed as a model multicomponent reaction (MCR) to efficiently prepare a library of polycondensates with continuously changed chain ...

Biginelli reaction starting directly from alcohols
Tetrahedron Letters, 2010Abstract An efficient of one-pot oxidative access to 3,4-dihydropyrimidin-2-(1H)-ones directly from aromatic alcohols under mild conditions is reported. The protocol involves 1-methylimidazolium hydrogen sulphate [Hmim]HSO4 catalyzed oxidation of aromatic alcohols to aromatic aldehydes with NaNO3 followed by their cyclocondensation with 1,3 ...

Recent Advances in Biginelli-type Reactions
Current Organic Chemistry, 2020The effective and high yielding synthesis of poly-functionalized pyrimidines, using multicomponent reactions (MCRs), is imperative in organic and medicinal chemistry. The classic Biginelli reaction is a typically one-pot three-component cyclocondensation reaction involving an aldehyde, a β-ketoester and urea, resulting in the construction of multi ...

The Biginelli Reaction Is a Urea-Catalyzed Organocatalytic Multicomponent Reaction
The Journal of Organic Chemistry, 2015The recently developed artificial force induced reaction (AFIR) method was applied to search systematically all possible multicomponent pathways for the Biginelli reaction mechanism. The most favorable pathway starts with the condensation of the urea and benzaldehyde, followed by the addition of ethyl acetoacetate.

A piperidinium triflate catalyzed Biginelli reaction
Tetrahedron, 2010A piperidinium triflate, 1,1,3,5-tetramethyl-4-oxo-2,6-diphenylpiperidinium triflate, in acetonitrile efficiently catalyzes one synthetic operational construction of biopertinent hydropyrimidines from respective aldehyde, β-dicarbonyl, and urea/thiourea building blocks.
