Viral‐Mediated Connexin 26 Expression Combined with Dexamethasone Rescues Hearing in a Conditional Gjb2 Null Mice Model

open access: yesAdvanced Science, EarlyView.
AAV2.7m8 serotype combined with the gfaABC1D promoter targets infection of supporting cells (SCs). AAV2.7m8‐gfaABC1D‐Gjb2 administration to mice results in excessive immune responses. The combination of AAV2.7m8‐gfaABC1D‐Gjb2 with dexamethasone (DEX) shows a synergistic effect and enhances the gene therapy effect in a conditional Cx26 null mice model ...

Clinical presentation, treatment, and outcome of 24 dogs with bacterial meningitis or meningoencephalitis without empyema (2010‐2020)

open access: yesJournal of Veterinary Internal Medicine, Volume 37, Issue 1, Page 223-229, January/February 2023., 2023
Abstract Background Bacterial meningitis (BM) and meningoencephalitis (BMEM) are associated with high case fatality rates and neurologic sequelae in people, but limited data exists on outcome in dogs. Hypothesis/Objectives To report the clinicopathologic features, treatment and outcome of BM/BMEM in dogs, with a focus on clinical presentation, relapse ...

Single Administration of AAV‐mAtp6v1b2 Gene Therapy Rescues Hearing and Vestibular Disorders Caused by Atp6v1b2‐Induced Lysosomal Dysfunction in Hair Cells

open access: yesAdvanced Science, EarlyView.
Wei et al. establish a hair cell‐specific conditional knockout mouse model (Atp6v1b2fl/fl;Atoh1Cre/+), and demonstrate the importance of Atp6v1b2 for hair cell through maintaining the survival of lysosomes. A single administration of AAV‐ie‐Eh3‐mAtp6v1b2 through scala media at P0‐P2 realizes function compensation and restores hearing and balance ...

Volume changes during active shape fluctuations in cells [PDF]

open access: yes, 2015
Cells modify their volume in response to changes in osmotic pressure but it is usually assumed that other active shape variations do not involve significant volume fluctuations. Here we report experiments demonstrating that water transport in and out of the cell is needed for the formation of blebs, commonly observed protrusions in the plasma membrane ...

Confinement by Liquid‐Liquid Interface Replicates In Vivo Neutrophil Deformations and Elicits Bleb‐Based Migration

open access: yesAdvanced Science, EarlyView.
Leukocytes sense mechanical cues provided by surrounding cells to regulate motility. Replicating the confining pressures applied by soft, elastic tissues in vitro is challenging. Here, the use of liquid‐liquid interfaces is introduced to model forces exerted by surrounding cells. Confining pressures are tunable by controlling interfacial curvature, and

Pyoderma Gangrenosum of the Hand: Unique Experience and Literature Review

open access: yesOrthopaedic Surgery, Volume 15, Issue 1, Page 362-365, January 2023., 2023
Pyoderma gangrenosum (PG) is usually a rare disease. However, knowledge of the disease is essential to actively consider PG at an early stage to help facilitate immediate treatment and avoid unnecessary interventions that may worsen the outcome. Background Pyoderma gangrenosum (PG) is a reactive, noninfectious, neutrophilic dermatosis.

Prevention of Ocular Scarring Post Glaucoma Filtration Surgery Using the Inflammatory Cell and Platelet Binding Modulator Saratin in a Rabbit Model [PDF]

open access: yes, 2012
Clinical Relevance: Late complications can occur with use of current antimetabolites to prevent scarring following glaucoma filtration surgery (GFS). Safer, more targeted, anti-fibrosis agents are sought. Objectives: The protein saratin has been shown to

Spontaneous contractility--mediated cortical flow generates cell migration in 3-dimensional environments [PDF]

open access: yes, 2010
We present a generic model of cell motility generated by acto-myosin contraction of the cell cortex. We identify analytically dynamical instabilities of the cortex and show that they trigger spontaneous cortical flows which in turn can induce cell migration in 3-dimensional (3D) environments as well as bleb formation.
