Total Neoadjuvant Therapy Versus Conventional Neoadjuvant Therapy for Rectal Cancer: Cost Analysis in a Public Healthcare System

open access: yesANZ Journal of Surgery, EarlyView.
Costs of neoadjuvant therapy. ABSTRACT Purpose Total neoadjuvant therapy (TNT) has become the standard of care in locally advanced rectal cancer, but its economic impact is unclear. This study compares the cost of TNT with conventional neoadjuvant therapy (CNT), consisting of either short‐course radiotherapy or long‐course chemoradiotherapy, within a ...

Phosphodiesterase 3 inhibitors boost bone outgrowth

open access: yesBritish Journal of Pharmacology, EarlyView.
Background and Purpose C‐type natriuretic peptide (CNP) stimulates skeletal growth by acting on the growth plates of long bones, and a CNP variant is clinically used for achondroplasia treatment. We previously reported that CNP stimulates the autonomic Ca2+ influx mediated by TRPM7 channels in growth plate chondrocytes to facilitate extracellular ...

Stabilisation of PRCP by deubiquitinase‐targeting chimera (DUBTAC) to replenish autophagy for ameliorating pathological cardiac hypertrophy

open access: yesBritish Journal of Pharmacology, EarlyView.
Background and Purpose Autophagy is essential for cellular homeostasis, and its impairment contributes to cardiac hypertrophy. Modulating autophagy has shown potential in treating pathological hypertrophy. Prolylcarboxypeptidase (PRCP), a lysosomal enzyme that hydrolyzes angiotensin II to Ang1‐7, has an unclear role in cardiac autophagy and hypertrophy.

Non‐canonical PKG1 regulation in cardiovascular health and disease

open access: yesBritish Journal of Pharmacology, EarlyView.
It is well established that the cyclic GMP‐dependent protein kinase I (PKG1) is canonically activated by cyclic guanosine monophosphate (cGMP), enabling its regulation of vascular tone, cardiac function and smooth muscle homeostasis. However, diverse non‐canonical stimuli of PKG1 have also been identified.

Role of Histone Deacetylase and Inhibitors in Cardiovascular Diseases

open access: yesCell Proliferation, EarlyView.
HDACs play an important role in the occurrence and development of cardiovascular diseases, such as myocardial hypertrophy, hypertension, and atherosclerosis. HDAC inhibitors have broad prospects for the treatment of CVD, and different HDAC subtypes can act through different mechanisms.
