Delivery of DNA‐encoded vaccines and proteins in mice using cutaneous suction‐mediated transfection
Cutaneous suction‐mediated transfection in mice for delivery of DNA‐encoded vaccines and proteins. A critical limitation of DNA vaccines and other therapeutics is transfection in vivo to produce the encoded antigens or therapeutic proteins. Cutaneous suction‐based methods have demonstrated effectiveness in many animal models and have been successfully ...

IMPLEMENTATION OF GENOME-WIDE SELECTION IN WHEAT
With the expected development of thousands of molecular markers in most crops, the marker-assisted selection theory has recently shifted from the use of a few markers targeted in QTL regions (or derived from candidate or validated genes) to the use of ...

We report the first β‐glucosidase with demonstrated hydrolytic activity on an N‐linked glycopyranoside. The enzyme, native to maize, was biochemically characterised for this novel reaction, and structural modelling of the enzyme–substrate complex revealed several clues to the underlying reduced catalytic rate relative to its native O‐glycopyranoside ...

Biophysical characterisation shows that NanX, a membrane transport protein from the major facilitator superfamily (MFS), forms both monomers and dimers after purification. AlphaFold modelling and substrate docking provide information on residues likely involved in substrate recognition for NanX and another MFS member, NanT.

We describe detailed protocols for the purification and preparation of Marchantia polymorpha Auxin Response Factor 2 (MpARF2). This protein is fused to an MBP solubility tag and an mNG fluorescent tag and is purified from Escherichia coli. The presented procedures make it possible to study MpARF2 assemblies, which could arise from phase separation ...

Breeding for Improved Nutritional Value
Abstract I consider myself to be a geneticist and plant breeder. Although not an expert in nutrition, I have long been interested in the improvement of nutritional value through plant breeding. This interest has led to a study of the genetic control of carotene synthesis.

The production and consumption of quinoa (Chenopodium quinoa Willd.) have grown rapidly in recent years, spreading far beyond its traditional growing region of the Andes Mountains in South America. The increase in consumption is due primarily to its high

Protocol for quantifying miRNA trafficking across the endosomal membrane
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.
