BTK mutation and ibrutinib resistance [PDF]

open access: yes, 2023
Provided herein are mutations in Bruton's Tyrosine Kinase (BTK) that confer resistance to treatment with a BTK inhibitors, such as Ibrutinib, and compositions and methods for the treatment, diagnosis, and characterization of BTK-inhibitor-resistant ...

Therapeutic effect of ibrutinib, a selective Bruton’s tyrosine kinase inhibitor, in orbital fibroblasts from patients with Graves’ orbitopathy

open access: yesPLoS ONE, 2022
Purpose Bruton’s tyrosine kinase (BTK) is an essential protein in B-cell antigen receptor (BCR) signaling pathway and is known to be related to pathogenetic effect on B-cell related malignancies and various autoimmune diseases.

Btk SH2-kinase interface is critical for allosteric kinase activation and its targeting inhibits B-cell neoplasms

open access: yesNature Communications, 2020
Constitutive Btk signaling drives several B-cell cancers. Here the authors demonstrate key allosteric intramolecular interactions between the SH2 domain and the kinase domain of Btk, and propose an alternative approach for inhibition of both wild-type ...

Phase II study of acalabrutinib in ibrutinib-intolerant patients with relapsed/refractory chronic lymphocytic leukemia

open access: yesHaematologica, 2021
B-cell receptor signalling inhibition by targeting Bruton tyrosine kinase (BTK) is effective in treating chronic lymphocytic leukemia (CLL). The BTK inhibitor ibrutinib may be intolerable for some patients.

Abstract 3232: Development of novel Btk inhibitor, CB988 targering ibrutinib-resistant Btk C481S mutant

open access: yes, 2017
Introduction: Bruton's tyrosine kinase (Btk) is a member of the Tec family of cytoplasmic tyrosine kinase. Btk plays a crucial role in the BCR signaling, essential for B-cell development, and Btk has been recognized as a validated therapeutic target for ...

Dasatinib inhibits CXCR4 signaling in chronic lymphocytic leukaemia cells and impairs migration towards CXCL12 [PDF]

open access: yes, 2012
Chemokines and their ligands play a critical role in enabling chronic lymphocytic leukaemia (CLL) cells access to protective microenvironmental niches within tissues, ultimately resulting in chemoresistance and relapse: disruption of these signaling ...

B Cells are Activated via a Nanoporous Interface that Stabilizes Microvilli and Engages Mechanosensitive Ion Channels

open access: yesAdvanced Science, EarlyView.
B cells can be activated independently of antigen recognition via mechanical stimulation through nanoporous substrates. Exposure to such substrates induced B cell microvilli extension into the pores, intracellular Ca2+ signaling, phosphorylation of signaling proteins, and CD69 expression.
