B cell linker protein (BLNK) is a regulator of Met receptor signaling and trafficking in non-small cell lung cancer

open access: yesiScience, 2022
Summary: Met is an oncogene aberrantly activated in multiple cancers. Therefore, to better understand Met biology and its role in disease we applied the Mammalian Membrane Two-Hybrid (MaMTH) to generate a targeted interactome map of its interactions with

Towards the First Principles in Biology and Cancer: New Vistas in Computational Systems Biology of Cancer [PDF]

open access: yesLife, 2021
These days many leading scientists argue for a new paradigm for cancer research and propose a complex systems-view of cancer supported by empirical evidence. As an example, Thea Newman (2021) has applied “the lessons learned from physical systems to a critique of reductionism in medical research, with an emphasis on cancer”.

B cell mechanobiology in health and disease: emerging techniques and insights into therapeutic responses

open access: yesFEBS Letters, EarlyView.
B cells sense external mechanical forces and convert them into biochemical signals through mechanotransduction. Understanding how malignant B cells respond to physical stimuli represents a groundbreaking area of research. This review examines the key mechano‐related molecules and pathways in B lymphocytes, highlights the most relevant techniques to ...

Genetic association and single-cell transcriptome analyses reveal distinct features connecting autoimmunity with cancers

open access: yesiScience, 2022
Summary: Autoimmune diseases (ADs) are at a significantly higher risk of cancers with unclear mechanism. By searching GWAS catalog database and Medline, susceptible genes for five common ADs, including systemic lupus erythematosus (SLE), rheumatoid ...

Hybrid Systems Modeling for (Cancer) Systems Biology [PDF]

open access: yes, 2015
AbstractThe advent of biological data of increasingly higher resolution in space and time has triggered the use of dynamic models to explain and predict the evolution of biological systems over space and time. Computer-aided system modeling and analysis in biology has led to many new discoveries and explanations that would otherwise be intractable to ...

In the pursuit of complexity: Systems medicine in cancer biology [PDF]

open access: yesCancer Cell, 2006
Adler et al., in a paper appearing in Nature Genetics, exploited the intersect of genetic information from expression profiles with that from array comparative genomic hybridization in human breast cancers to identify genes that may induce the transcription of the prognostic "wound response" expression signature. The amplification of two genes, MYC and

Assessing the carcinogenic potential of low-dose exposures to chemical mixtures in the environment: the challenge ahead. [PDF]

open access: yes, 2015
Lifestyle factors are responsible for a considerable portion of cancer incidence worldwide, but credible estimates from the World Health Organization and the International Agency for Research on Cancer (IARC) suggest that the fraction of cancers ...

The thioredoxin‐like and one glutaredoxin domain are required to rescue the iron‐starvation phenotype of HeLa GLRX3 knock out cells

open access: yesFEBS Letters, EarlyView.
Glutaredoxin (Grx) 3 proteins contain a thioredoxin domain and one to three class II Grx domains. These proteins play a crucial role in iron homeostasis in eukaryotic cells. In human Grx3, at least one of the two Grx domains, together with the thioredoxin domain, is essential for its function in iron metabolism.

Multi-omics analysis reveals neoantigen-independent immune cell infiltration in copy-number driven cancers

open access: yesNature Communications, 2018
Neoantigen load has been associated with tumour immune infiltration. Here, the authors show that while this is true for tumours with recurrent mutations, cancers with recurrent CNAs show neoantigen-independent infiltration driven by cytokine production ...

Evolutionary interplay between viruses and R‐loops

open access: yesFEBS Letters, EarlyView.
Viruses interact with specialized nucleic acid structures called R‐loops to influence host transcription, epigenetic states, latency, and immune evasion. This Perspective examines the roles of R‐loops in viral replication, integration, and silencing, and how viruses co‐opt or avoid these structures.
