Cardiotonic agents. 2. (Imidazolyl)aroylimidazolones, highly potent and selective positive inotropic agents

Journal of Medicinal Chemistry, 1987
A series of 4-alkyl-1,3-dihydro-5-[(1H-imidazolyl)benzoyl]-2H-imidazol-2-ones 9 was synthesized and evaluated in vitro for positive inotropic and cyclic AMP phosphodiesterase inhibitory activity. A wide range of inotropic and enzyme-inhibitory potencies was observed, substitution on the imidazolyl moiety being the major determinant of activity.

Cardiotonic agents in the management of chronic cardiac failure

American Heart Journal, 1982
Heart failure is a syndrome with distinct clinical signs and symptoms. The severity of cardiac failure and a deterioration in functional capacity can be determined by a progressive exercise test and by the noninvasive determination of maximum oxygen uptake.

Cardiovascular Properties of a New Cardiotonic Agent, MDL 19205

Journal of Cardiovascular Pharmacology, 1984
The cardiovascular properties of a new cardiotonic agent, MDL 19205, were investigated in anesthetized and conscious dogs and in a dog heart-lung preparation. MDL 19205 (0.1-1 mg/kg), administered to anesthetized dogs by intravenous injection, produced a dose-related increase in cardiac contractile force lasting up to 1 h. It also produced a relatively

[Membranotropic action of cardiotonic agents and ubiquinone].

Ukrainskii biokhimicheskii zhurnal (1978), 1991
Interaction of cardiotonic drugs (strophantidine acetate, suphan, para-oxybenzoic acid) and ubiquinone with phospholipid bilayers has been studied. Exothermic effect of the reaction followed by an increase in microviscosity and hydrophobicity of the bilayer from cardiolipin, but by a decrease of the microviscosity of the bilayer from lecithin has been ...

Calcium Signaling and Pharmacology of Cardiotonic Agents

1995
The mechanisms of action of drugs that act on myocardial contractility through different signal transduction pathways have lately been elucidated to a great extent, in particular since the introduction of novel methods for detecting changes in the intracellular concentration of Ca++ ions, ([Ca++] i), by the application of aequorin, quin-2, fura-2 and ...

Inosine : a naturally occurring cardiotonic agent.

Journal de pharmacologie, 1984
For many years, Inosine was considered to be a simple metabolite of adenosine which was devoid of any cardiovascular effects. This theoretical ineffectiveness can be explained in the light of recent studies by the use of inadequate doses. In fact, higher doses of inosine, a non-toxic nucleoside, have demonstrated, experimentally, a cardiovascular ...

[New cardiotonic/inodilator agents: energetic aspects].

Zeitschrift fur Kardiologie, 1993
Inotropic agents alter myocardial oxygen consumption by influencing heart rate, by influencing preload and afterload due to vasodilation, and by direct effects on the myocardium. The latter critically depend on the pharmacologic mode of action. Inotropic agents which act by increasing cyclic AMP in the failing human myocardium increase myocardial ...

[Value and limitations of new cardiotonic agents].

Bulletin de l'Academie nationale de medecine, 1989
New cardiotonic agents are an original approach in the treatment of severe heart failure. They can be classified into two groups: --beta-adrenergic agonists which stimulate beta-adrenergic receptors and, therefore, increase cyclic AMP production and intracellular calcium concentration; --phosphodiesterase inhibitors which block the cyclic AMP ...

Inhibition of some new cardiotonic agents by tetrodotoxin.

Arzneimittel-Forschung, 1987
The effects of sulmazole, milrinone and 1,5-dihydro-6-chloro-3-methylimidazo[2,1-b]quinazolone-2 on guinea pig left atria were studied, measuring the force of contraction in the absence and in the presence of 1 x 10(-5) mol/l octahydro-12-(hydroxymethyl)-2-imino-5,9: 7,10a-dimethano-10aH-[1,3]-dioxocino[6,5-d]pyrimidine- 4,7,10, 11,12-pentol ...
