Synthesis of Milrinone, a Cardiotonic Agent

A new synthesis of milrinone from 6-methyl-5-(4-pyridinyl)-2(1H)-pyridone is reported. A convenient synthesis of 3,4'-bipyridines is achieved. They can be converted into pyridone by hydrogenolysis and hydrolysis, respectively.

The effect on the contractile protein system of MCI-154, a novel and potent cardiotonic agent, was examined by using thin bundles of chemically skinned fibers from the guinea-pig papillary muscles.
