セファロスポリンCは,2,4-ジニトロフェニルヒドラジンの過塩素酸溶液と沸騰水浴中で加熱すると,黄色のグリオキサール・ビス2,4-ジニトロフェニルヒドラゾンを生成するから,これを濾取し,テトラメチルアンモニウムヒドロキサイド含有のジメチルホルムアミドで溶解すると,λmaxが615mμにある青色となり,2×10-8~2×10-7mol/mlの範囲でベールの法則を満足し,定量に利用できる.本定量法において,グリオキサールの生成部位が,セファロスポリン母核の6,7位の炭素からであるので,セファロスポリン類および,類似構造のペニシリン類の定量にも利用できることがわかった.
