火烈鸟性别鉴定的分子标记方法

由于火烈鸟在外观上雌雄极为相似,很难用常规方法对其性别进行鉴定。本试验首次确立了利用PCR技术对火烈鸟性别进行鉴定的分子标记方法。从火烈鸟血液中提取总基因组DNA,针对性染色体上的CHD基因和EE0.6序列,筛选了4对引物进行特异性扩增,其中3对引物组合成2个组(A/B和B/C)进行多重PCR。结果表明,3组引物都在雄性个体中扩增出1条片段,在雌性中扩增出2条片段;待测群体中雄性个体11只,雌性个体6只。经检验,利用3组引物都能准确鉴定火烈鸟性别,为该物种在分子水平上的性别鉴定奠定了基础。

PCR技术在麦哲伦企鹅性别鉴定上的应用

以青岛水族馆7只麦哲伦企鹅的新鲜血液为对象提取基因组DNA,根据基因组数据设计性别特异引物P2/P8,对其CHD基因进行PCR扩增,回收扩增的目的片段进行测序,获得这些麦哲伦企鹅的性别相关基因序列。将这7只麦哲伦企鹅的CHD基因序列与GenBank上公布的短趾雕的CHD基因序列进行同源性比对分析,建立遗传进化树,初步确定编号为1572、1573、1574、1575、36的麦哲伦企鹅鉴定为雄性,编号29和1577号麦哲伦企鹅为雌性。研究表明,雌雄麦哲伦企鹅CHD基因的内含子基因序列存在差异,P2 ...

一种准确简便的东方白鹳性别分子鉴定方法

东方白鹳为国家I级重点保护动物,在自然条件下繁殖率很低,进行迁地保护及再引入措施对遗传多样性保护具有举足轻重的作用,成功地进行性别鉴定是其笼养人工繁殖的前提。本研究对4只东方白鹳,运用CHD基因的一对引物2550F/2718R进行了准确简便的性别鉴定,并对其序列结合其他5种远缘鸟类相应序列进行分析,证实了此种方法的科学性和可信度,分析了性别鉴定机理。这种安全、方便、准确的性别分子鉴定方法将有助于东方白鹳迁地保护及再引入措施的实施。通过分析可以看出,此种方法未来存在着广阔的应用前景。

朱鹮性别分子鉴定的研究进展

朱鹮(Nipponia nippon)为世界濒危物种,也是国家一级重点保护鸟类。虽然一系列的保护措施显著增加了朱鹮种群规模和分布区域,但近交衰退、低繁殖力和雏鸟高致残率等问题一直制约着朱鹮种群的发展。朱鹮由于雌雄同型,难以从外观区分性别,因此性别鉴定将对其人工繁殖具有重要意义。本文综述了朱鹮性别的分子鉴定方法,主要包括基于性染色体连锁的染色体解旋酶DNA结合蛋白CHD基因及EcoRⅠ片段的PCR鉴定方法,以期进行有效的人工繁育工作,实现对朱鹮种群的保护及规模重建。
