The alkylating properties of chlorambucil
Pharmacology Biochemistry and Behavior, 1994Previous work has indicated an aziridinium ion mechanism in the hydrolysis of chlorambucil, and the present work on the alkylation of nucleophiles fully supports this mechanism. This mechanism forms the basis for understanding the kinetics of alkylation reactions because their rates are limited by the rate of formation of the aziridinium ion and the ...

The comparative toxicity of chlorambucil and chlorambucil-spermidine conjugate to BALBc mice
Cancer Letters, 1994The acute intraperitoneal toxicities of chlorambucil and chlorambucil-spermidine conjugate have been compared, in mice. Both compounds were neurotoxic and also caused a prolonged fall in bodyweight and a depletion of lymphocyte numbers associated with a fall in the total leukocyte count and loss of spleen and thymus weight. Alanine aminotransferase and

Degradation of Chlorambucil in Aqueous Solution
Journal of Pharmaceutical Sciences, 1980The stability of chlorambucil and its degradation product 4-[p-(2-chloroethyl-2-hydroxyethylamino)phenyl]butyric acid (I) was studied using reversed-phase high-pressure liquid chromatography. The degradation rate of chlorambucil was unaffected by pH between pH 5 and 10 but decreased at lower pH. The degradation rates of chlorambucil and I differed only

Interaction of chlorambucil with tRNA
Canadian Journal of Biochemistry, 1978Preincubation of purified mixed tRNAs from Escherichia coli K12-MO with 2.94 mM chlorambucil (CAB) for 2 h at 37 °C results in the inhibition of the capacity of mixed tRNAs to accept alanine, arginine, asparagine, aspartic acid, glutamic acid, glutamine, glycine, histidine, isoieucine, leucine, lysine, methionine, phenylalanine, proline, serine ...

Cutaneous Reaction to Chlorambucil
Archives of Dermatology, 1977To the Editor.— Chlorambucil is an oral alkylating agent that has been used since early 1950, particularly for the treatment of chronic lymphatic leukemia. The side effects are gastrointestinal symptoms, such as nausea and vomiting, and bone marrow depression. Allergic skin reactions are extremely rare.

Chlorambucil and Acute Intermittent Porphyria
Clinical Oncology, 2002A 54-year-old women with quiescent acute intermittent porphyria (AIP) developed low-grade non-Hodgkin's lymphoma. She was successfully treated with chlorambucil without any significant rise in urinary proto-porphyrins. The case is described and a review of chemotherapy use in AIP is presented.

Study of Chlorambucil and Chlorambucil–Trimethyl-β-cyclodextrin Inclusion Complex by CE
Chromatographia, 2007The results of CE studies of solid inclusion complexes of chlorambucil (CHL) and trimethyl-β-cyclodextrin (TM-β-CD) are presented. Chlorambucil has low stability in aqueous solution, so the formation of complexes with β-CD is a good way to improve its stability and dissolution. The experimental results confirm the existence of inclusion complexes.

The pharmacokinetics of prednimustine and chlorambucil in the rat
Cancer Chemotherapy and Pharmacology, 1981In the rat prednimustine, the prednisolone ester of chlorambucil, is much less toxic than equimolar doses of chlorambucil, when administered subcutaneously (SC). This is due to differences in alkylating agent pharmacokinetics. Prednimustine injected SC produced low plasma concentrations (less than 5 microM) of the alkylating metabolites chlorambucil ...

A Binding Site for Chlorambucil on Metallothionein
Biochemistry, 1996It is of interest to test the hypothesis that induced metallothionein (MT) acts in acquired drug resistance by covalent sequestration. In this study MT was incubated in vitro with chlorambucil (CHB) under conditions where only 1:1 covalent adducts were formed.
