Expression and purification of human cholesterol 7 alpha-hydroxylase in Escherichia coli.

open access: yesJournal of Lipid Research, 1994
Cholesterol 7 alpha-hydroxylase (P450c7) is the first and rate-limiting enzyme in bile acid biosynthesis and is the product of a cytochrome P450 gene, CYP7.

Hormonal regulation of the cholesterol 7 alpha-hydroxylase gene (CYP7)

open access: yesJournal of Lipid Research, 1995
The transcriptional regulation of the rat cholesterol 7 alpha-hydroxylase gene (CYP7) by hormones and signal transduction pathways was studied by transient transfection assay of the promoter activity. HepG2 cells were transfected with deletion mutants of

Endotoxin, TNF, and IL-1 decrease cholesterol 7 alpha-hydroxylase mRNA levels and activity.

open access: yesJournal of Lipid Research, 1996
Endotoxin (LPS) and cytokines increase cholesterol synthesis and the secretion of lipoproteins by the liver in rodents resulting in hypercholesterolemia.

Bile acid excretion and cholesterol 7 alpha-hydroxylase expression in hypercholesterolemia-resistant rabbits

open access: yesJournal of Lipid Research, 1993
We have developed a partially inbred substrain of New Zealand white rabbits (CRT/mlo) that are resistant to the hypercholesterolemia that accompanies cholesterol feeding to normal rabbits.

Measurement of cholesterol 7 alpha-hydroxylase activity with selected ion monitoring.

open access: yesJournal of Lipid Research, 1981
Formation of 7 alpha-hydroxycholesterol by rat liver microsomes was quantitated using a gas chromatograph-mass spectrometer operated in selected ion monitoring mode.

Effect of dietary cholesterol and taurocholate on cholesterol 7 alpha-hydroxylase and hepatic LDL receptors in inbred mice

open access: yesJournal of Lipid Research, 1993
Compared to BALB/c mice, inbred C57BL/6 mice are more susceptible to developing fatty streak atherosclerotic lesions when fed a cholesterol-rich diet containing taurocholate. We examined the metabolic basis for the taurocholate requirement.

Cholesterol 7 alpha-hydroxylase from human liver: partial purification and reconstruction into defined phospholipid-cholesterol vesicles.

open access: yesJournal of Lipid Research, 1981
Cholesterol 7 alpha-hydroxylase, the rate-limiting enzyme for bile acid synthesis, was shown to be copurified with human liver microsomal cytochrome P-450.

Dietary fats modulate the regulatory potential of dietary cholesterol on cholesterol 7 alpha-hydroxylase gene expression

open access: yesJournal of Lipid Research, 1997
Cholesterol 7 alpha-hydroxylase (cyp7) is the rate-limiting enzyme in bile acid biosynthesis. Previously, dietary cholesterol was shown to induce cyp7 gene expression. However, recent studies have produced data that are inconsistent with this observation,

Oxysterol 7α-Hydroxylase Activity by Cholesterol 7α-Hydroxylase (CYP7A) [PDF]

open access: yesJournal of Biological Chemistry, 2000
A 7 alpha-hydroxylation is necessary for conversion of both cholesterol and 27-hydroxycholesterol into bile acids. According to current theories, cholesterol 7 alpha-hydroxylase (CYP7A) is responsible for the former and oxysterol 7 alpha-hydroxylase (CYP7B) for the latter reaction.
