Targeted metabolomics reveals novel diagnostic biomarkers for colorectal cancer

open access: yesMolecular Oncology, EarlyView.
This study employed targeted metabolomic profiling to identify 302 distinct metabolites present in platelet‐rich plasma (PRP), revealing aberrant metabolic profiles amongst individuals diagnosed with colorectal cancer (CRC). Compared to carcinoembryonic antigen (CEA) and cancer antigen 19‐9 (CA199), our metabolite panel showed improved sensitivity ...

Retrieval-Reasoning Large Language Model-based Synthetic Clinical Trial Generation [PDF]

open access: yesarXiv
Machine learning (ML) exhibits promise in the clinical domain. However, it is constrained by data scarcity and ethical considerations, as the generation of clinical trials presents significant challenges due to stringent privacy regulations, high costs, and the extended duration required for conducting studies with human participants.

Adversity Index for Clinical Trials: An Inclusive Approach for Analysis of Safety Data [PDF]

open access: yesarXiv, 2018
This paper proposes a new method for analysis of adverse event data in clinical trials. The method is illustrated by application to data on 4 phase III clinical trials, two on breast cancer and two on diabetes mellitus.

KMT2A degradation is observed in decitabine‐responsive acute lymphoblastic leukemia cells

open access: yesMolecular Oncology, EarlyView.
We demonstrate that decitabine (DEC) not only degrades the DNA methyltransferase DNMT1 but also the leukemic driver lysine methyltransferase KMT2A likely due to structural similarity of the DNA‐binding CXXC domains. DEC influences KMT2A downstream processes and synergizes with menin inhibitor revumenib (REV) to decrease leukemic cell proliferation, and

From Bench to Bedside: A Review of Clinical Trials in Drug Discovery and Development [PDF]

open access: yesarXiv
Clinical trials are an indispensable part of the drug development process, bridging the gap between basic research and clinical application. During the development of new drugs, clinical trials are used not only to evaluate the safety and efficacy of the drug but also to explore its dosage, treatment regimens, and potential side effects.

Polyfunctional CD8+CD226+RUNX2hi effector T cells are diminished in advanced stages of chronic lymphocytic leukemia

open access: yesMolecular Oncology, EarlyView.
CD226+CD8+ T cells express elevated levels of RUNX2, exhibit higher proliferation capacity, cytokines and cytolytic molecules expression, and migratory capacity. In contrast, CD226−CD8+ T cells display an exhausted phenotype associated with the increased expression of co‐inhibitory receptors and impaired effector functions.

Inhibition of acyl‐CoA synthetase long‐chain isozymes decreases multiple myeloma cell proliferation and causes mitochondrial dysfunction

open access: yesMolecular Oncology, EarlyView.
Triacsin C inhibition of the acyl‐CoA synthetase long chain (ACSL) family decreases multiple myeloma cell survival, proliferation, mitochondrial respiration, and membrane potential. Made with Biorender.com. Multiple myeloma (MM) is an incurable cancer of plasma cells with a 5‐year survival rate of 59%.
