Bile‐Derived Exosomal miR‐196a/‐196b as Diagnostic Biomarkers Associated With Tumor Progression in Biliary Tract Cancer

Bile‐derived exosomal miR‐196a and miR‐196b are identified as novel biomarkers for biliary tract cancer. Serum miR‐196a levels are significantly elevated in advanced disease with metastasis, while both miRNAs contribute to tumor progression by promoting cancer cell proliferation and invasion.

KYNU in Gastric Cancer Cells Promotes Tumor Progression by Influencing Macrophage Polarization Via PF4

KYNU is upregulated in gastric cancer cells and activates the PI3K/Akt pathway, leading to PF4 secretion. PF4 drives nonclassical macrophage polarization with high expression of S100A8 and MMP7, which promotes tumor proliferation, migration, and invasion while suppressing phagocytosis, thereby accelerating gastric cancer progression.

Genetic Evolution in BRCA1‐Associated Breast Cancer Reveals Early Driver Mutations Shaping Tumor Features and Prognosis

Multi‐region sequencing of 57 BRCA1‐associated breast cancers identified TP53 as the dominant initial driver, defining a triple‐negative subgroup with biallelic BRCA1 loss and elevated genomic instability. TP53 truncating mutations were enriched in BRCA1 carriers and linked to reduced HRD, EMT activation, and a trend toward worse survival.

Collecting duct carcinoma of the kidney

Six cases of renal epithelial neoplasms, which show atypical histological features previously described in cases of collecting duct carcinomas, are presented. The tumours were of a tubulo‐papillary architecture, arose in the renal medulla and showed prominent infiltrating tubules with an associated desmoplastic reaction.

Collecting duct carcinoma is an unusual variant of renal cell carcinoma, whose appearance and behavior are not well established. We identified six cases of collecting duct carcinoma in our files. The clinical, pathologic, and immunohistochemical characteristics of these tumors are reported. The most common symptom was gross hematuria (four cases).

Collecting duct carcinoma (Bellini duct carcinoma) is a very rare disease with poor prognosis. Primary tumor surgery is not advisable, even though it is the therapy of choice for suspected malignant space-occupying renal masses. Chemotherapy seems to be inferior to new therapeutic concepts based on single case reports.
