Real-space and real-time dynamics of CRISPR-Cas9 visualized by high-speed atomic force microscopy

open access: yesNature Communications, 2017
CRISPR RNA-guided endonuclease Cas9 recognizes and cleaves the double-stranded DNA complementary to the RNA guide. Here the authors use high-speed atomic force micropcopy (HS-AFM) to visualize the conformational dynamics of Cas9 during its DNA targeting ...

Twist versus Nonlinear Stacking in Short DNA Molecules

open access: yes, 2014
The denaturation of the double helix is a template for fundamental biological functions such as replication and transcription involving the formation of local fluctuational openings.

Multidimensional OMICs reveal ARID1A orchestrated control of DNA damage, splicing, and cell cycle in normal‐like and malignant urothelial cells

open access: yesMolecular Oncology, EarlyView.
Loss of the frequently mutated chromatin remodeler ARID1A, a subunit of the SWI/SNF cBAF complex, results in less open chromatin, alternative splicing, and the failure to stop cells from progressing through the cell cycle after DNA damage in bladder (cancer) cells. Created in BioRender. Epigenetic regulators, such as the SWI/SNF complex, with important

Metal-mediated base pairs in parallel-stranded DNA

open access: yesBeilstein Journal of Organic Chemistry, 2017
In nucleic acid chemistry, metal-mediated base pairs represent a versatile method for the site-specific introduction of metal-based functionality.

Escape from TGF‐β‐induced senescence promotes aggressive hallmarks in epithelial hepatocellular carcinoma cells

open access: yesMolecular Oncology, EarlyView.
Chronic TGF‐β exposure drives epithelial HCC cells from a senescent state to a TGF‐β resistant mesenchymal phenotype. This transition is characterized by the loss of Smad3‐mediated signaling, escape from senescence, enhanced invasiveness and metastatic potential, and upregulation of key resistance modulators such as MARK1 and GRM8, ultimately promoting

Cloning of DNA complementary to ornithine aminotransferase mRNA.

open access: yesJournal of Biological Chemistry, 1982
Free polysomes were isolated from the livers of rats maintained on a 60% casein diet to induce ornithine aminotransferase mRNA. Ornithine aminotransferase-synthesizing polysomes were immunoadsorbed using monospecific, affinity-purified antibody and Staphylococcus aureus cells.
