From Batch to Continuous: Freeze-Drying of Suspended Vials for Pharmaceuticals in Unit-Doses
Industrial & Engineering Chemistry Research, 2019In response to the current trend in the pharmaceutical industry, a new concept for the freeze-drying of pharmaceuticals in unit-doses is presented: the continuous freeze-drying/lyophilization of su...

Investigation of Continuous and Intermittent Heating on Far-Infrared Assisted Freeze-Drying
Transactions of the ASABE, 2009Freeze-drying of sweet potato was studied using continuous and intermittent far-infrared (FIR) assisted heating in this study. Intermittency levels of a = 0, 0.25, 0.5, 0.75, and 1 were applied with a cycle time of 30 min, and it was found that the optimum intermittency level was a = 0.5.

Near-infrared chemical imaging (NIR-CI) is an emerging tool for process monitoring because it combines the chemical selectivity of vibrational spectroscopy with spatial information. Whereas traditional near-infrared spectroscopy is an attractive technique for water content determination and solid-state investigation of lyophilized products, chemical ...

The use of a novel microbalance (Christ) technique to monitor continuously the weight loss of a vial standing on a shelf of a freeze-dryer has been investigated. The drying rates of the following aqueous solutions were measured during the primary drying phase of a complete freeze-drying cycle: sucrose (75 mg/mL, 2.5-mL fill volume), sucrose and ...

Recently, a continuous freeze-drying process for the production of unit doses was presented and evaluated. In this concept, the freezing step is modified compared to traditional batch freeze-drying, as glass vials filled with a liquid formulation, are rotated around their longitudinal axis while cooled and frozen with a cold, sterile and inert gas (i.e.

The continuous precipitation of zinc carbonates using aqueous solutions of zinc nitrate and sodium carbonate was quenched within a few seconds by combining a micromixer with a bench-top spray dryer. In this way, it was possible to monitor the slow phase transformation of the initially formed sodium zinc carbonate into zinc hydroxy carbonate during ...

Freeze-drying of suspended vials: a first step toward continuous manufacturing
2016The new paradigm of pharmaceutical industry is to move from batch to continuous processes in order to satisfy the stringent requirements of quality, safety and efficiency set by regulatory authorities and reduce production costs. In this perspective, freeze-drying needs to be completely rethought in order to be more integrated in the chain of ...

Present study aimed to determine to what extent freeze-dried spermatozoa were able to withstand high-temperature conditions: transient increase in storage temperature and long-term exposure to room temperature. Mouse spermatozoa were freeze-dried in EGTA/Tris-HCl buffered solution alkalinized using KOH (K-ETBS, pH 7.7), and then stored for up to 7 ...
