Evaluation of a PAT-based in-line control system for a continuous spin freeze-drying process

International Journal of Pharmaceutics, 2023
Continuous spin freeze-drying provides a range of opportunities regarding the implementation of several in-line process analytical technologies (PAT) to control and optimize the freeze-drying process at the individual vial level. In this work, two methods were developed to (1) control the freezing phase by separately controlling the cooling and ...

Investigation of Continuous and Intermittent Heating on Far-Infrared Assisted Freeze-Drying

Transactions of the ASABE, 2009
Freeze-drying of sweet potato was studied using continuous and intermittent far-infrared (FIR) assisted heating in this study. Intermittency levels of a = 0, 0.25, 0.5, 0.75, and 1 were applied with a cycle time of 30 min, and it was found that the optimum intermittency level was a = 0.5.

Potential of Near-Infrared Chemical Imaging as Process Analytical Technology Tool for Continuous Freeze-Drying

Analytical Chemistry, 2018
Near-infrared chemical imaging (NIR-CI) is an emerging tool for process monitoring because it combines the chemical selectivity of vibrational spectroscopy with spatial information. Whereas traditional near-infrared spectroscopy is an attractive technique for water content determination and solid-state investigation of lyophilized products, chemical ...

The Impact of Formulation Composition and Process Settings of Traditional Batch Versus Continuous Freeze-Drying On Protein Aggregation

Journal of Pharmaceutical Sciences, 2020
The long-term stability of therapeutic protein products can be extended by freeze-drying. However, the freeze-drying process itself has several harmful stresses. A rationalized formulation design can significantly mitigate protein damage caused by freezing, dehydration and interfacial stresses of lyophilization and reconstitution.

Evaluation of immersion‐contact type heat transfer for continuous pharmaceutical spin freeze‐drying process

Journal of Food Process Engineering, 2019
AbstractSpin freeze‐drying process is a recently proposed method to achieve continuous processing of unit doses in freeze‐drying application. An experimental study is carried out to investigate the heat transfer and drying characteristics of a spin‐frozen liquid product.

Developing a framework to model the primary drying step of a continuous freeze-drying process based on infrared radiation

European Journal of Pharmaceutics and Biopharmaceutics, 2018
The continuous freeze-drying concept based on spinning the vials during freezing and on non-contact energy transfer via infrared (IR) radiation during drying, improves process efficiency and product quality (uniformity) compared to conventional batch freeze-drying. Automated control of this process requires the fundamental mechanistic modelling of each

Continuous measurement of drying rate of crystalline and amorphous systems during freeze‐drying using an in situ microbalance technique

Journal of Pharmaceutical Sciences, 2001
The use of a novel microbalance (Christ) technique to monitor continuously the weight loss of a vial standing on a shelf of a freeze-dryer has been investigated. The drying rates of the following aqueous solutions were measured during the primary drying phase of a complete freeze-drying cycle: sucrose (75 mg/mL, 2.5-mL fill volume), sucrose and ...

Dual chamber cartridges in a continuous pharmaceutical freeze-drying concept: Determination of the optimal dynamic infrared heater temperature during primary drying

International Journal of Pharmaceutics, 2019
The applicability of DCCs in a continuous freeze-drying concept based on spin freezing and infrared heating was evaluated. Maximum applicable filling volume was evaluated. Secondly the mechanistic model for the determination of the optimal dynamic infrared heater temperature during primary drying of regular vials during continuous freeze-drying was ...
