Proteasomal degradation of intracellularly expressed Amblyomin‐X limits suicide gene therapy potential in melanoma cells

open access: yesFEBS Open Bio, EarlyView.
This study explores the feasibility of expressing the antitumoral protein Amblyomin‐X through a suicide gene therapy approach and investigates its intracellular fate after gene delivery. Although the gene is efficiently expressed, melanoma cells rapidly degrade the Amblyomin‐X protein via proteasome activity.

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Fatigue crack growth. Overload and underload interactions [PDF]

open access: yes, 2006
This article deals with the study of the overload- underload interactions effects on fatigue crack growth (FCG) rate. With the purpose of understanding of overload-underload interaction mechanisms the FEM modeling of stress-strain state at the crack tip ...

YIPFα1A expression is regulated by multilayered molecular mechanisms

open access: yesFEBS Open Bio, EarlyView.
YIPFα1A, a five‐pass Golgi protein, is regulated at multiple layers. (1) Rare‐codon enrichment drives translation‐coupled mRNA decay. (2) A proximal 3′‐UTR element stabilizes mRNA. (3) A distal 3′‐UTR element included by alternate poly(A) site usage represses translation, which can be overridden by the proximal 3′‐UTR element.

Observation and analysis of process zone–crack interaction in crack layer during fatigue crack propagation in polypropylene

open access: yesJournal of Materials Research and Technology
Fatigue crack propagation in polypropylene, despite its significance in a wide range of structural applications, has not been sufficiently researched because of the complex interaction between the crack tip and process zone ahead of the crack tip.

Sudden anaerobization in Amphibacillus xylanus increases intracellular labile ferrous iron and inhibits cell growth

open access: yesFEBS Open Bio, EarlyView.
Abruptly changing from aerobic to anaerobic conditions (sudden anaerobization) induced growth inhibition and a significant increase in intracellular labile ferrous iron in the aerotolerant anaerobe Amphibacillus xylanus. We found that free flavins mediate efficient electron transfer from NADH to ferric iron under anaerobic conditions, suggesting that ...

Effect of rolling on fatigue crack growth rate of Wire and Arc Additive Manufacture (WAAM) processed Titanium [PDF]

open access: yes, 2013
Titanium (Ti) alloys have been commonly used in the aerospace industry, not only because they have a high strength-to-weight ratio (comparing to the steels) but also their satisfactory corrosion resistance.

Versatile vector tools for efficient protein screening across multiple expression systems

open access: yesFEBS Open Bio, EarlyView.
A unified vector toolkit enables rapid protein expression screening across E. coli, insect, and mammalian cells. A single primer pair amplifies the target gene, which is inserted into any vector via a standardized interface. This streamlined workflow eliminates repeated cloning steps, accelerating the identification of optimal expression conditions for

On the cyclic J-integral of a 3D semi elliptical surface crack

open access: yes, 2014
This paper investigates an approach for calculating the cyclic J-Integral (ΔJ) through a new industrial application. A previously proposed method is investigated further with the extension of this technique through a new application of a practical 3D ...

The crystal structure of the Borrelia burgdorferi nicotinamidase BBE22 resolves a long‐standing annotation error

open access: yesFEBS Open Bio, EarlyView.
The crystal structure of Borrelia burgdorferi nicotinamidase (PncA/BBE22) reveals the correct full‐length protein initiated from a non‐canonical AUU start codon. The structure validates previous biochemical findings and resolves a long‐standing annotation error, demonstrating that the truncated database sequence is structurally incompatible with the ...
