The effects of 5-fluorodeoxyuridine show that an impairment of DNA synthesis increases crossing-over.

Secondary crossing-over between alleles seems to arise as a consequence of modifications of the normal crossover mechanism caused by a particular mutation. Interference between crossovers is believed to be polarized in relation to the centromere, but secondary crossovers are thought not to interfere with one another.

Cytogenetic and Genome Research, 1967Experiments with human lymphocytes and H.Ep.2 human neoplastic cells treated with tritiated thymidine yielded evidence which suggests that sister chromatid crossing-over may occur in the centromere. As a result of this study a possible mechanism of isochromosome formation is postulated.
