Methyl CpG binding protein 2 regulates CYP27A1-induced myometrial contraction during preterm labor.
Molecular human reproductionPersistent and intense uterine contraction is a risk factor for preterm labor. We previously found that Methyl-CpG binding protein 2 (MeCP2), as a target of infection-related microRNA miR-212-3p, may play an inhibitory role in regulating myometrium ...

Role of CYP27A1 in progesterone metabolism in vitro and in vivo
American Journal of Physiology-Endocrinology and Metabolism, 2009In the kidney, progesterone is inactivated to 20α-dihydro-progesterone (20α-DH-progesterone) to protect the mineralocorticoid receptor from progesterone excess. In an attempt to clone the enzyme with 20α-hydroxysteroid activity using expression cloning in CHOP cells and a human kidney expression library, serendipitously cDNA encoding CYP27A1 was ...

Three siblings with Cerebrotendinous Xanthomatosis: A novel mutation in the CYP27A1 gene
European Journal of Medical Genetics, 2012Cerebrotendinous Xanthomatosis (CTX) is a rare autosomal recessive lipid storage disease caused by sterol 27-hydroxylase (CYP27) deficiency. We report three CTX siblings that shared a novel mutation of the CYP27A1 gene. These siblings presented with elevated cholestanol levels and typical manifestations such as tendon xanthomas, cataracts, osteopenia ...

Studies on sterol 27-hydroxylase (CYP27A1)
2004Human sterol 27-hydroxylase (CYP27A1) has been studied in the present thesis with focus on substrate specificity and possible role of the enzyme for prevention of accumulation of cholesterol and cholestanol in tissues. Attempts have been made to evaluate a metabolic pathway to bile acids in human primary hepatocytes that starts with a 27-hydroxylation ...

Glucocorticoid receptor-mediated upregulation of human CYP27A1, a potential anti-atherogenic enzyme
Biochimica et Biophysica Acta (BBA) - Molecular and Cell Biology of Lipids, 2008Sterol 27-hydroxylase (CYP27A1) is required for the hepatic conversion of cholesterol into bile acids and for production of 27-hydroxycholesterol which affects cholesterol homeostasis in several ways. Dexamethasone increases hepatic bile acid biosynthesis and CYP27A1-mediated enzyme activity in HepG2 cells.

A novel mutation in the CYP27A1 gene in a family with cerebrotendinous xanthomatosis
International Journal of Neuroscience, 2020Cerebrotendinous xanthomatosis (CTX) is an inherited disorder associated with abnormal deposition of cholestenol in the brain and other tissues. Here we report a Chinese family with two affected members of CTX.Clinical data were collected. Gene analysis, MRI, neuropsychological assessments, and the biopsy of right Achilles tendon xanthoma were carried ...

β‐Cryptoxanthin uptake in THP‐1 macrophages upregulates the CYP27A1 signaling pathway
Molecular Nutrition & Food Research, 2013ScopeMitochondrial sterol 27‐hydroxylase (CYP27A1), a mediator of cholesterol homeostasis, is reported to exhibit antiatherogenic properties. Many studies suggested that all‐transretinoic acid can be used to treat atherosclerosis through retinoic acid receptor (RAR)‐mediated upregulation of CYP27A1 expression.

Mast cells express CYP27A1 and CYP27B1 in epithelial skin cancers and psoriasis
European Journal of Dermatology, 2015Ultraviolet (UV) radiation and the vitamin D system are involved in immunosuppression in the skin. Previous in vitro and animal studies suggest a role for mast cells in these mechanisms. To study vitamin D3 metabolizing enzymes, CYP27A1 and CYP27B1, in mast cells in epithelial skin cancers and psoriasis.Biopsies were collected from the non-lesional and ...

Regulation of human CYP27A1 by estrogens and androgens in HepG2 and prostate cells
Archives of Biochemistry and Biophysics, 2007The regulation of the human CYP27A1 gene by estrogens and androgens was studied in human liver-derived HepG2 and prostate cells. Our results show that the promoter activity, enzymatic activity and mRNA levels of CYP27A1 in HepG2 cells are downregulated by estrogen in presence of ERalpha or ERbeta.
