HERV‐K TM Subunit Elicits CD8+ T Cell Anergy and Tumor Immune Evasion via Targeting CD3 Coreceptor ε in AML and PDAC

Novel viral immune checkpoint K‐TM subunit is aberrantly expressed in CD8+ T cells and enriched in sera of cancer patients, inducing CD8+ T cell anergy and eliciting potent tumor immune escape in mouse model. Mechanistically, K‐TM directly targets and induces the phosphorylation of CD3ε receptor.

Spatial transcriptomic analysis of Anaplastic thyroid carcinoma (ATC)‐differentiated thyroid carcinoma (DTC) coexisting tumors reveals that coexisting DTC regions with ATC‐like genomic alterations undergo sequential transcriptomic reprogramming and immune microenvironment remodeling, ultimately evolving into a full ATC pathological phenotype.

METTL10‐Mediated PIAS3 Methylation Links Purine Metabolism to Gastric Cancer Progression

METTL10‐mediated methylation of PIAS3 at K442 inhibits its SUMO E3 ligase activity toward MITF. This reduces MITF SUMOylation and subsequent RNF4‐mediated ubiquitination, leading to MITF protein accumulation and gastric cancer progression.
