Drugs that act on both G protein‐coupled receptors (GPCRs) and kinases: potentiation of effects, side effects and general aspects of drug pleiotropy

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background A drug designed for a specific target often interacts with multiple targets, either unintentionally or as part of its intended mechanism of action. This has been called pharmacological pleiotropy or polypharmacology. There are key endogenous ligands such as ATP, GABA and glutamate that act on various proteins in humans. Furthermore,

Amylin analogues as a novel perspective in anti‐obesity therapy

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Public health systems face a major challenge because of the worldwide obesity epidemic. The number of obese individuals has increased dramatically and, even in Switzerland which may be considered a relatively “lean” country, the numbers are alarmingly high and have reached almost 50% of the adult population.

AKR1C3 Binds β‐Trcp to Promote the Degradation of TFRC to Protect Hepatocellular Carcinoma From Ferroptosis

open access: yesCancer Science, EarlyView.
In this study, we show that aldo‐keto reductase family 1 member C3 (AKR1C3), a protein highly expressed in ferroptosis resistant hepatocellular carcinoma cells, acts as an enzyme‐independent scaffold protein by promoting the nuclear export of β‐transducin repeat protein (β‐TrCP) and its binding to transferrin receptor (TFRC) in an enzyme‐independent ...

Diverging in Vivo Immune Response Patterns After Oral Food Challenge in Peanut Allergic Adults

open access: yesClinical &Experimental Allergy, EarlyView.
After the oral food challenge, peanut (PN)‐allergic adults were monitored over 4 weeks, which revealed diverging immune response patterns, with strong responders (SR) exhibiting heightened Ara h 2+ B cell expansion, PN sIgE induction and increasing PN‐specific Tconv. Weak responders (WR) had more activated PN‐specific Tregs.
