Patient therapy outcome modeling in cancer organoids is improved by cancer‐associated fibroblasts and organoid assembly convolution

open access: yesMolecular Oncology, EarlyView.
Patient‐derived organoids (PDOs) from pancreatic, colorectal, and gastric cancers were used to evaluate standard and experimental therapies. Incorporating cancer‐associated fibroblasts (CAFs) into organoid cultures improved patient therapy outcome prediction.

Critical conversations: a user-centric approach to chatbots for history taking in the pediatric intensive care unit

open access: yesFrontiers in Pediatrics
In this article, we describe the potential utility and design of chatbots to improve history taking in the pediatric intensive care unit (PICU). The fast-paced, high-stakes environment of the PICU often forces clinicians to obtain only enough information

ZW4864‐mediated inhibition of the β‐catenin/BCL9/BCL9L complex reveals therapeutic potential in bladder cancer

open access: yesMolecular Oncology, EarlyView.
BCL9 and BCL9L drive bladder cancer progression by enhancing β‐catenin signaling, promoting proliferation, migration, invasion, and organoid growth. Genetic depletion of BCL9(L) suppresses malignant phenotypes, while pharmacological disruption of the β‐catenin/BCL9(L) complex with ZW4864 inhibits canonical Wnt signaling and tumor‐associated cellular ...

Modelling of diagnostic reasoning [PDF]

open access: yes, 1989
Medical diagnosis can be modelled in terms of the classical notions of abduction, deduction and induction. Abduction is making a preliminary guess that allows to establish a set of plausible diagnostic hypotheses, followed by deduction for exploring ...

Systemic dysregulation of apolipoproteins in amyotrophic lateral sclerosis serum

open access: yesFEBS Open Bio, EarlyView.
Amyotrophic lateral sclerosis (ALS) is a fatal disease that damages motor neurons. This study found that people with ALS show significant changes in blood fats and the proteins that carry them. Several apolipoproteins were higher, lipid balances were altered, and normal protein–lipid relationships were disrupted.

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Using script theory to cultivate illness script formation and clinical reasoning in health professions education

open access: yesCanadian Medical Education Journal, 2015
Background: Script theory proposes an explanation for how information is stored in and retrieved from the human mind to influence individuals’ interpretation of events in the world.

Long‐term hippocampal alterations and cognitive impairment in a murine model of surgical sepsis

open access: yesFEBS Open Bio, EarlyView.
Using a mouse model of surgical sepsis, we tested long‐term memory and analyzed the transcriptome of single cells isolated from the hippocampus. Survivor mice showed worse memory, loss of certain brain cell subpopulations, and abnormal immune cell activity—suggesting that post‐sepsis brain alterations may be linked to cognitive deficits.

Premature conclusions in diagnostic reasoning

open access: yesAcademic Medicine, 1985
The purpose of the study reported here was to explore the characteristics of premature diagnostic conclusions in a group of physicians, medical students, and residents. When the subjects were asked to construct complete, precise problem lists from three case abstracts, premature closure occurred frequently, it could be recognized with good interrater ...
