Mycobacterial cell division arrest and smooth‐to‐rough envelope transition using CRISPRi‐mediated genetic repression systems

open access: yesFEBS Open Bio, EarlyView.
CRISPRI‐mediated gene silencing and phenotypic exploration in nontuberculous mycobacteria. In this Research Protocol, we describe approaches to control, monitor, and quantitatively assess CRISPRI‐mediated gene silencing in M. smegmatis and M. abscessus model organisms.

Design of Low-Power Structural FIR Filter Using Data-Driven Clock Gating and Multibit Flip-Flops

open access: yesJournal of Electrical and Computer Engineering, 2020
Optimization for power is one of the most important design objectives in modern digital signal processing (DSP) applications. The digital finite duration impulse response (FIR) filter is considered to be one of the most essential components of DSP, and ...

Promiscuous stimulation of HSP70 ATPase activity by parasite‐derived J‐domains

open access: yesFEBS Open Bio, EarlyView.
The malaria parasite Plasmodium falciparum exports three highly homologous yet functionally divergent J‐domain proteins into human erythrocytes. Here, we show that J‐domains isolated from all three proteins effectively stimulate the ATPase activity of both endogenous host and exported parasite HSP70 chaperones.

Applicability of mitotic figure counting by deep learning: a development and pan‐cancer validation study

open access: yesFEBS Open Bio, EarlyView.
In this study, we developed a deep learning method for mitotic figure counting in H&E‐stained whole‐slide images and evaluated its prognostic impact in 13 external validation cohorts from seven different cancer types. Patients with more mitotic figures per mm2 had significantly worse patient outcome in all the studied cancer types except colorectal ...

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

The Design Of Intelligent Filter For EEG

open access: yesMATEC Web of Conferences, 2018
EEG data processing method is usually digital filter designed by the traditional method. Its disadvantage is the transition zone is wide and the filtering effect is poor.

Area- and Power-Efficient Nearly-Linear Phase Response IIR Filter by Iterative Convex Optimization

open access: yesIEEE Access, 2019
Low complexity infinite impulse response (IIR) filter design with nearly-linear phase response has attracted considerable attention in recent years due to the substantially high area and power consumption of linear phase finite impulse response (FIR ...

Design of multi-decimation rate digital filter for sigma-delta ADC

open access: yesDianzi Jishu Yingyong, 2022
Based on the standard 0.18 μm process, a digital decimation filter applied to the Sigma-Delta analog-to-digital converter is designed, which can change the decimation rate and adapt to different signal bandwidths. The filter adopts multi-stage decimation

Development of human monoclonal antibodies against TARM1 by yeast display

open access: yesFEBS Open Bio, EarlyView.
Human monoclonal antibodies against TARM1 are generated by yeast display‐guided selection. These antibodies bind to soluble and cell‐surface forms of TARM1. Also, these antibodies exhibit agonistic activity in the NFAT‐GFP reporter assay, indicating that TARM1 signaling can be functionally modulated by antibodies and suggesting TARM1 as a potential ...

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,
