PFKFB4 Deubiquitination by USP10 Enhances Fumarate Metabolism to Orchestrate the KDM1A/Rad51 Axis and Confer Radioresistance in Lung Cancer

USP10 binds to and stabilizes PFKFB4, enhancing glycolytic ATP production, which activates the urea cycle and elevates fumarate. This inhibits histone demethylase KDM1A, leading to increased H3K4me1 enrichment at the Rad51 promoter and direct activation of Rad51 transcription, which confers lung cancer radioresistance. Resistance training combined with ursolic acid supplementation restores anabolic signaling (PI3K/mTORC1) suppressed by diabetes, helping preserve muscle mass. This strategy may offer a potential therapeutic approach to combat muscle atrophy and support healthy aging in diabetic conditions.

Biomarkers of Sarcopenia: Current Status and Future Perspectives

In this review, we summarize the various biomarkers discovered in recent years, including biochemical, imaging, and physical testing markers. We analyze their advantages and disadvantages and propose potential ways to improve the diagnostic accuracy of biomarkers, as well as future research directions.

ABSTRACT In patients with Smith–Magenis syndrome (SMS), an inverted circadian rhythm of melatonin (MT) contributes to the sleep disturbance. Standard treatment of sleep disturbance with MT often leads to extremely high daytime MT levels, resulting in even more sleep disorders. We therefore retrospectively evaluated the MT data of 89 SMS patients.

ABSTRACT Autosomal recessive HARS1‐related disorder (originally described as Usher syndrome type 3B) caused by a homozygous Y454S variant in the histidyl‐tRNA synthetase gene (HARS1) is characterized by progressive sensorineural hearing and vision loss and respiratory deterioration with risk for sudden death following febrile illnesses.
