Doxycycline and Hepatotoxicity

The hepatotoxicity of tetracyclines is well known. If microvesicular steatosis due to a high dose of tetracycline has virtually disappeared, it can also be observed with other drugs belonging to the tetracycline family. To our knowledge, hepatotoxicity induced by doxycycline has never been reported.

Doxycycline Therapy for Leptospirosis

To study antibiotic efficacy, 29 patients with leptospirosis were treated in a randomized, double-blinded fashion with doxycycline, 100 mg orally twice a day, or placebo. Therapy was given for 7 days in a hospital, and patients were followed for 3 weeks afterwards. Duration of illness before therapy and severity of illness were the same in both groups.

Little is known about the behaviour of antibiotics in the tissues. To investigate this problem, rats were sacrificed in groups of six, 1, 2, 4, 6, 8, 12 and 18 h after an intraperitoneal injection of 10 mg doxycycline/kg body weight. The antibiotic levels were determined in the nine major organs and in the serum by a microbiological method.

The Disposition of Doxycycline by the Rat

The disposition by rats of tritium-labeled doxycycline was investigated. More drug is eliminated from the circulatory system by diffusion into the lumen of the small intestine than by excretion with urine or bile. Doxycycline Prophylaxis for Shigellosis
