β‐TrCP overexpression enhances cisplatin sensitivity by depleting BRCA1

open access: yesMolecular Oncology, EarlyView.
Low levels of β‐TrCP (Panel A) allow the accumulation of BRCA1 and CtIP, which facilitate the repair of cisplatin‐induced DNA damage via homologous recombination (HR) and promote tumor cell survival. In contrast, high β‐TrCP expression (Panel B) leads to BRCA1 and CtIP degradation, impairing HR repair, resulting in persistent DNA damage and apoptosis ...

DRUG ANTAGONISM AND AUDIOGENIC SEIZURES IN MICE [PDF]

open access: yesBritish Journal of Pharmacology and Chemotherapy, 1959
The occurrence of audiogenic seizures in mice has been used as the basis of a method for measuring the antagonism between sodium thiopentone and bemegride or leptazol. Thiopentone in doses of 8 and 16 mg./kg. protected all the mice against the occurrence of audiogenic seizures.

Antagonism between Two Mechanisms of Antifungal Drug Resistance [PDF]

open access: yesEukaryotic Cell, 2006
ABSTRACT This study tested for interaction between two independently evolved mechanisms of fluconazole resistance in Saccharomyces cerevisiae . One set of strains was from a 400-generation evolution experiment, during which the concentration of fluconazole was increased from 16 to 256 μg/ml in four ...

Community assessment to advance computational prediction of cancer drug combinations in a pharmacogenomic screen [PDF]

open access: yes, 2019
The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal ...

Adenosine‐to‐inosine editing of miR‐200b‐3p is associated with the progression of high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
A‐to‐I editing of miRNAs, particularly miR‐200b‐3p, contributes to HGSOC progression by enhancing cancer cell proliferation, migration and 3D growth. The edited form is linked to poorer patient survival and the identification of novel molecular targets.

Suppression of bacterial cell death underlies the antagonistic interaction between ciprofloxacin and tetracycline

open access: yesMolecular Systems Biology
Antibiotic combinations aim to maximise drug treatment efficiency and minimise resistance evolution, but a full understanding of their effect on bacterial cells is lacking.

Antagonism of a PCP Drug Discrimination by Hallucinogens and Related Drugs [PDF]

open access: yesNeuropsychopharmacology, 2000
Drugs such as PCP and MK-801 can cause psychotic reactions in humans by antagonizing NMDA receptors. This action is ultimately toxic to certain cortical neurons and may be one mechanism underlying neurodegenerative diseases, including schizophrenia. It has been reported that hallucinogens such as LSD, DOM, and DOI can block the neurotoxic effects of ...

Inhibition of CDK9 enhances AML cell death induced by combined venetoclax and azacitidine

open access: yesMolecular Oncology, EarlyView.
The CDK9 inhibitor AZD4573 downregulates c‐MYC and MCL‐1 to induce death of cytarabine (AraC)‐resistant AML cells. This enhances VEN + AZA‐induced cell death significantly more than any combination of two of the three drugs in AraC‐resistant AML cells.

Research and development of 2-ethyl-6-methyl-3-oxypyridine succinate nano-form for the treatment of epilepsy

open access: yesЭпилепсия и пароксизмальные состояния, 2019
The aim is to develop an antiepileptic drug based on polymer nanoparticles with 2-ethyl-6-methyl-3-oxypyridine succinate to facilitate the drug transport through the blood-brain barrier.Materials and methods.

Differentiated effects of the multimodal antidepressant vortioxetine on sleep architecture: Part 1, a pharmacokinetic/pharmacodynamic comparison with paroxetine in healthy men [PDF]

open access: yes, 2015
We compared the effect of vortioxetine, paroxetine and placebo after three days of dosing on sleep architecture. This was a randomised, double-blind, four-way crossover, placebo-controlled, multiple-dose study in 24 healthy young men.
